基于GWAS数据库鉴定与食管癌相关的易感基因 被引量:2

摘要 目的:寻找并鉴定与食管癌相关的遗传易感基因。方法:通过搜索全基因组关联研究(genome-wide association studies,GWAS)数据库下载与食管癌相关的单核苷酸多态性(single nucleotide polymorphism,SNP)位点,利用全基因组表达定量性状位点(genome-wide expression quantitative trait locus,eQTL)数据库对SNPs进行靶基因预测。选择在以上eQTL数据库中得到两次验证的基因在食管癌肿瘤组织、癌旁组织和正常组织中进行检测,通过基因表达的差异分析确定与食管癌相关基因。结果:GWAS数据库中共发现40个SNPs与食管癌具有关联。在4个eQTL数据库中共发现20个SNPs参与调节34个基因的调控,其中eQTL数据库中得到两次验证的基因为丝氨酸羧化酶基因(serine racemase,SRR)、卷曲螺旋结构域117基因(coiled-coil domain containing 117,CCDC117)、细胞周期检测点激酶2(checkpoint kinase 2,CHEK2)、X-盒结合蛋白1(X-box binding protein 1,XBP1)和NOC3样DNA复制调节因子(NOC3 like DNA replication regulator,NOC3L)。食管癌肿瘤组织中CCDC117(t=6.25,P<0.001,差异倍数=1.90),CHEK2(t=9.37,P<0.001,差异倍数=3.08),XBP1(t=10.77,P<0.001,差异倍数=2.67)呈现高表达,肿瘤组织与癌旁组织在CHEK2(t=2.17,P=0.03)和XBP1(t=1.86,P<0.001)的表达差异有统计学意义。结论:在GWAS数据库中发现多个与食管癌相关的SNPs位点,通过eQTL数据库分析和外周血检测发现这些SNPs可能通过参与调控基因XBP1、CCDC117和CHEK2进而影响食管癌的发生。
