摘要
目的探讨转化生长因子-β1(TGF-β1)对人皮肤鳞状细胞癌A431细胞增殖的影响及可能的机制。方法将HaCaT细胞和人皮肤鳞状细胞癌A431细胞在不同浓度及时间的TGF-β1作用后,采用MTT法检测其增殖情况,RT-PCR法检测其TGF-β受体mRNA表达水平的变化。结果不同浓度的TGF-β1均能显著抑制HaCaT细胞的增殖,与对照组比较差异有统计学意义(P<0.05),且随着TGF-β1浓度的升高,其抑制作用逐渐增强;但在A431细胞中,不同浓度的TGF-β1组与对照组之间差异无统计学意义(P>0.05)。用相同浓度的TGF-β1分别作用不同时间后,HaCaT细胞的增殖受到明显抑制,与对照组比较差异有统计学意义(P<0.05)。但在A431细胞中,其抑制作用不明显,与对照组比较差异无统计学意义(P>0.05)。RT-PCR结果显示,随着TGF-β1浓度的升高,A431细胞中TGF-β受体ⅠmRNA(TGF-βRⅠmRNA)表达逐渐下降。相同浓度TGF-β1作用下,TGF-βRⅠmRNA的表达在一定时间范围内呈时间依赖性下调趋势。结论皮肤鳞状细胞癌A431细胞对TGF-β1引起的生长抑制不敏感,这可能是通过调控TGF-β1受体表达水平所致。
Objective To investigate the effect of transforming growth factor-β1(TGF-β1)on the proliferation of human squamous cell carcinoma A431 cells and its possible mechanism.Methods HaCaT cells and skin squamous cell carcinoma A431 cells were treated with TGF-β1 at different concentrations and time.The proliferation of HaCaT cell and A431 cell was detected by MTT assay.The expression of TGF-βreceptor mRNA was detected by RT-PCR.Results Different concentrations of TGF-β1 could significantly inhibit the proliferation of HaCaT cell(P<0.05),and the inhibitory effect increased gradually with the increases of TGF-β1 concentration.However,in the skin squamous cell carcinoma A431 cell group,there was no significant difference between the different concentrations of TGF-β1 group and the control group(P>0.05).After the same concentration of TGF-β1 was applied for different time,the proliferation of HaCaT cells was significantly inhibited,and the difference was statistically significant(P<0.05).The inhibition was positively correlated with time in a certain time range.However,in the skin squamous cell carcinoma A431 cell group,the inhibitory effect was not obvious,and there was no significant difference(P>0.05).The results of RT-PCR showed that with the increase of TGF-β1 concentration,the expression of TGF-βreceptorⅠmRNA(TGF-βRⅠmRNA)in skin squamous cell carcinoma A431 cells and control cells decreased gradually.After the same concentration of TGF-β1 was applied for different time,the change of TGF-βRⅠmRNA had showed a time-dependent downward trend.Conclusion Skin squamous cell carcinoma A431 cells are not sensitive to growth inhibition by TGF-β1,which may be caused by regulation of TGF-β1 receptor expression levels.
