摘要
目的:探究微小RNA-34b-5p(miR-34b-5p)、微小RNA-150(miR-150)在内蒙古地区蒙古族哮喘患者外周血单个核细胞(PBMC)中的表达水平及意义。方法:选取2013年1月~2014年1月本院收治的120例蒙古族哮喘患者为蒙古哮喘组,以同期本院就诊的汉族哮喘患者120例为汉族哮喘组,并选择同期健康体检者120例为健康组;RT-PCR法检查所有研究对象PBMC中miR-34b-5p、miR-150表达水平;肺功能仪检测各组肺功能指标;ELISA检测各组受试者外周血中IFN-γ、IL-2、IL-4、IL-13的水平;分析上述指标与疾病严重程度的相关性;评价miR-34b-5p、miR-150表达水平对哮喘的诊断价值。结果:汉族哮喘组和蒙古哮喘组患者miR-34b-5p、IFN-γ、IL-2水平及FEV1%均明显低于健康组,miR-150、IL-4及IL-13显著高于健康组(P<0.05);蒙古族和汉族患者IFN-γ、IL-2水平及FEV1%比较无显著差异(P>0.05);蒙古族miR-34b-5p显著低于汉族;而miR-150、IL-4及IL-13则显著高于汉族(P<0.05);miR-150为疾病严重程度的保护因素(OR=0.081,P=0.023),而miR-34b-5p为疾病严重程度的危险因素(OR=20.898,P=0.015);两族患者开展联合检测的诊断效能均显著优于miR-34b-5p及miR-150单一检测(z=12.257,10.947,P<0.001)。结论:miR-34b-5p及miR-150与内蒙古地区哮喘患者疾病严重程度显著相关,且不同种族之间哮喘患者表达水平差异明显,二者联用能对哮喘诊断的效果具有显著提升,并且可用于评价患者疾病的严重程度。
Objective:To explore the expression level and significance of microRNA-34b-5p(miR-34b-5p)and microRNA-150(miR-150)in peripheral blood mononuclear cells(PBMC)of Mongolian asthmatic patients in Inner Mongolia.Methods:120 cases of Mongolian asthma patients admitted to our hospital from January 2013 to January 2014 were selected as Mongolian asthma group,120 cases of Han asthma patients in our hospital during the same period were selected as Han asthma group,and 120 cases of health persons were selected as health group.The expression levels of miR-34b-5p and miR-150 in PBMC of all subjects were examined by RT-PCR.Pulmonary function test for each group of lung function indicators.The levels of IFN-γ,IL-2,IL-4 and IL-13 in peripheral blood of each group of subjects were detected by ELISA.To analyze the correlation between the above indicators and the severity of the disease;to evaluate the diagnostic value of miR-34b-5p and miR-150 expression levels in asthma.Results:The levels of miR-34b-5p,IFN-γ,IL-2 and FEV1%in the Han asthma group and the Mongolian asthma group were significantly lower than those in the healthy group,and miR-150,IL-4 and IL-13 were significantly higher than the healthy group(P<0.05).There were no significant differences in IFN-γ,IL-2 levels and FEV1%between Mongolian and Han patients(P>0.05).Mongolian miR-34b-5p was significantly lower than Han nationality;miR-150,IL-4 and IL-13 were significantly higher than Han nationality(P<0.05).miR-150 was a protective factor for disease severity(OR=0.081,P=0.023),while miR-34b-5p was a risk factor for disease severity(OR=20.898,P=0.015).The diagnostic efficacy of the combined detection of both patients was significantly better than that of miR-34b-5p and miR-150(z=12.257,10.947,P<0.001).Conclusion:miR-34b-5p and miR-150 were significantly associated with the severity of asthma in Inner Mongolia,and the expression levels of asthmatic patients were significantly different between different ethnic groups.The combination of the two can significantly improve the effectiveness of asthma diagnosis,and can be used to assess the severity of the patient′s disease.
