生物信息学技术预警番泻叶用于乳腺癌便秘患者的用药风险 被引量:3

摘要 目的基于生物信息学技术,探讨番泻叶对于乳腺癌便秘患者的可能作用机制及潜在不良反应,预警相关用药风险,为临床合理用药研究提供参考。方法采用生物信息学技术,从高通量基因表达数据库中下载基因表达谱数据芯片GSE85871,筛选番泻叶泻下主要成分番泻苷A干预人乳腺癌细胞MCF7后的差异表达基因,并对其进行基因本体论数据库功能注释、京都基因与基因组百科全书数据库通路富集分析、蛋白网络互作分析。结果番泻叶应用于乳腺癌便秘患者时可能导致结肠黑变病,伴随中枢性食欲减弱及细胞间信息传递、糖代谢、炎症反应、心肌收缩和情绪异常,可能影响乳腺癌的发生与发展。结论应用番泻叶时应密切关注该类患者的消化、神经、免疫、心血管系统及糖代谢相关机体状态,并在使用过程中持续关注患者原患疾病乳腺癌的发展情况。 Objective To explore the potential adverse reactions of Sennae Folium in constipation patients with breast cancer based on the bioinformatics and warn the risks so as to provide reference for the research on rational use of drugs in constipation patients with breast cancer.Methods Methods of bioinformatics were employed.The microarray data of GSE85871 was downloaded from GEO database,and analyzed by means of gene ontology annotation,Kyoto Encyclopedia of Genes and Genomes pathway analysis and protein-protein interaction networks.Results The analysis showed that when used in constipation patients with breast cancer,Sennae Folium might lead to melanosis coli,along with abnormalities of appetite,cell signaling,carbohydrate metabolism,inflammatory response,myocardial contraction and mood,thus affecting the development of breast cancer.Conclusion It is recommended that close attention should be paid to the body state of patients related to digestive,nervous,immune,cardiovascular systems and carbohydrate metabolism.Clinicians should keep track of the development of patients'preexisting disease.
