类叶牡丹皂苷G在大鼠体内的组织分布研究 被引量:1

摘要 目的:建立了一种UPLC-MS/MS法,测定类叶牡丹皂苷G(Cauloside G)在大鼠体内各主要脏器(心、肝、脾、肾、脑)中的分布特点及组织药物浓度随时间变化的动态规律。方法:大鼠灌胃给予Cauloside G150 mg/kg,给药后0.5、1、2、4和6 h 5个时间点,收集心、肝、脾、肾、脑组织样品,处理后通过三重四级杆线性离子阱串联质谱仪分析。采用ACQUITY UPLC HSS T3(2.1 mm×150 mm,1.8μm)色谱柱,流动相0.1%甲酸水溶液-0.1%甲酸乙腈溶液,梯度洗脱,流速0.3 mL·min~(-1),电喷雾离子源,负离子模式,多重反应监测(MRM)。结果:大鼠灌胃给予Cauloside G后,Cauloside G在大鼠的心、肝、脾、肾和脑中均有分布。心脏组织中,Cauloside G是在给药0.5 h后浓度达到最高值,浓度为(53.7±7.60)ng/mL;肝、脾、肾、脑组织中,Cauloside G是给药1 h后浓度达到峰值,浓度分别为(90.1±6.19)ng/mL,(103.2±17.67)ng/mL,(101.8±14.32)ng/mL,(68.2±14.76)ng/mL。结论:Cauloside G在大鼠体内的各个组织均有分布,其中肝、脾、肾组织中药物浓度较高,该研究为类叶牡丹中三萜皂苷类成分的进一步开发和利用提供参考。 Objective:To detect the distribution of Cauloside G in the main organs of the rats,including the heart,liver,spleen,kidney and brain,and to study the dynamic changes of drug concentration with time by UPLC-MS/MS.Methods:Rats were intragastrically administrated with 150 mg/kg Cauloside G.At the time of 0.5,1,2,4 and 6 h,the heart,liver,spleen,kidney and brain tissues of the rats were collected,and they were analyzed by UPLC-MS.The chromatography separation was performed on a ACQUITY UPLC HSS T3 column(2.1 mm×150 mm,1.8μm)with 0.1%formic acid aqueous solution and 0.1%formic acid acetonitrile solution for gradient elution(0.3 mL·min-1).Heated electrospray ionization source was applied in a negative ion mode for multiple reaction monitoring(MRM).Results:After intragastrical administration of Cauloside G,it was detected to be distributed in rat heart,liver,spleen,kidney and brain.The concentration of Cauloside G in rat heart peaked at 0.5 h(53.7±7.60)ng/mL.In addition,its concentration in liver(90.1±6.19)ng/mL,spleen(103.2±17.67)ng/mL,kidney(101.8±14.32)ng/mL and brain(68.2±14.76)ng/mL peaked at 1 h.Conclusion:Cauloside G is distributed in the main organs of the rats.Its concentration remains high in liver,spleen and kidney.The results provided references for the development and the application of triterpenoid saponins from Caulophyllum robustum Maxim.
