摘要
从鱼腥草中提取出一种水溶性多糖,运用化学分析法、光谱学法、色谱法及扫描电镜法对鱼腥草多糖(Houttuynia cordata polysaccharide,HCP)的基本理化性质、相对分子量、单糖组成及固态形貌等进行分析并对其体外抗氧化活性进行研究。结果显示HCP为含吡喃糖结构的酸性果胶类均一多糖,其重均相对分子量为3.87×105 Da,且该多糖由鼠李糖、阿拉伯糖、半乳糖、葡萄糖、木糖、甘露糖、半乳糖醛酸、葡萄糖醛酸8种单糖组成。扫描电镜分析发现HCP主要由杆状、丝状和球状物质组成。体外抗氧化实验表明HCP有明显的DPPH自由基和·OH的清除作用,其清除活性可分别高达到87.18%和31.37%。因此,本研究的结果可为HCP的生物活性研究奠定基础,且有利于鱼腥草资源的开发应用。
The aim of this work was to isolated a water-soluble polysaccharide(HCP)from Houttuynia cordata Thunb and to analyze its basic physicochemical properties,relative molecular weight,monosaccharide composition,solid state morphology and vitro antioxidant activity by chemical analysis,spectroscopy,chromatography and scanning electron microscopy.Our results showed that HCP was an acid pectin polysaccharide with pyranose structure,and its weight average relative molecular weight was 3.87×105 Da.Furthermore,HCP was composed of eight monosaccharides including rhamnose,arabinose,galactose,glucose,xylose,mannose,galacturonic acid and glucuronic acid.Also,the images of scanning electron microscopy indicated that the rod-shaped,filiform and spherical substances were shown in HCP.Moreover,the data of antioxidant experiments indicated that HCP displayed significant scavenging effects against DPPH radicals and·OH in vitro and their scavenging activities were 87.18%and 31.37%respectively.Therefore,these findings were beneficial to providing a foundation for the biological research activity of HCP and promoting the development and utilization of Houttuynia cordata Thunb resources.
