摘要
以聚-L-谷氨酸大分子为原料,通过酸水解反应使其生成聚合度较低的聚谷氨酸片段,然后用pH敏感的肼基在小分子聚谷氨酸末端连接疏水性抗肿瘤药多柔比星,并使其自组装成纳米胶束,同时利用包埋作用将多柔比星负载到胶束中,该胶束载药量高,且可利用细胞内p H值作为触发将其靶向肿瘤细胞,希望为靶向抗癌药物的研究提供基础和参考。
Poly-L-glutamic acid(PLGA) macromolecules were used as raw materials to form poly(L-glutamic acid) fragments with low degree of polymerization by acid hydrolysis. Then, the hydrophobic antitumor drug doxorubicin was connected connected to the end of the small molecule of PLGA by pH sensitive hydrazine group, and then the copolymer was self-assembled into nano micelles,which were high drug load and pH sensitivity.This study is expected to provide basis and reference for the research of targeted anticancer drugs.
