摘要
优化双刺参胶囊中总皂苷的提取工艺,建立总皂苷含量的测定方法,并考察提取物的体外抗氧化活性。采用紫外分光光度法,确定总皂苷的含量测定方法并对其进行方法学考察;采用单因素试验与正交试验相结合的方法,以总皂苷提取率为指标优化提取工艺。结果显示:采用香草醛-乙醇-硫酸显色法测定双刺参胶囊总皂苷具有较好的专属性,齐墩果酸在4.51~31.56μg/mL质量浓度范围内线性关系良好(R^2=0.9994),平均加样回收率为102.19%,RSD为1.40%(n=9),可以用以检测双刺参胶囊中的总皂苷含量;在最佳提取工艺条件下,双刺参胶囊总皂苷提取率为28.84 mg/g;通过对清除ABTS自由基阳离子和抗脂质过氧化作用的考察,结果发现双刺参提取物具有一定的抗氧化能力,但弱于VC。结果表明双刺参胶囊总皂苷的提取工艺提取效率高、稳定可行;建立的总皂苷含量测定方法具有良好的精密度、重复性,结果准确可靠,为双刺参胶囊的下一步开发和质量评估提供了科学依据。
The extraction process of total saponins from Shuangcishen capsules was optimized,the determination method of total saponins content was established,and the antioxidant activity of the extracts in vitro was also investigated.The content of total saponin was determinized by UV spectrophotometry method and its methodology was investigated.The combination method of single factor experiment and orthogonal experiment was applied to optimize the extraction process using total saponins extraction rate as index of evaluation.The results showed that the total saponins of Shuangcishen capsules had good specificity by vanillin-ethanol-sulfuric acid colorimetry method.Oleanolic acid had a good linear relationship between 4.51μg/mL and 31.56μg/mL(R^2=0.9994).The recovery rate of the sample was 102.19%,and the RSD was 1.40%(n=9),which can be used to detect the total saponins content in Shuangcishen capsules.Under the optimal extraction conditions,the total saponins extraction rate of Shuangcishen capsules could reach 28.84 mg/g.The extracts of Shuangcishen had certain antioxidant activity,but weaker than that of VC according to the investigation of both ABTS free radical cation scavenging and anti-lipid peroxidation.The results indicated that the extraction process of the total saponins of Shuangcishen capsules was high,stable and feasible,and the determination method of total saponins content had good precision and repeatability.The research results were accurate,reliable and reproducible,which provided a scientific basis to carry further development and quality assessment to Shuangcishen capsules.
