摘要 目的通过Box-Behnken法优化天麻的冻干工艺,为其工业化生产提供理论依据。方法通过 Box-Behnken 响应面法,以浸出物含量、天麻素含量、对羟基苯甲醇含量及巴利森苷类含量的综合评分作为评价指标,考察预冷冻时间、干燥温度、一次干燥时间3个因素对天麻真空冷冻干燥工艺的影响,优化天麻的冻干工艺条件。结果确定天麻的最佳冻干工艺条件为预冻时间10 h,一次干燥时间18 h,干燥温度45℃。结论优化的天麻冻干工艺具有重复性和可操作性,可用于工业化生产。 Objective To optimize the freezing-drying process for Gastrodiae Rhizoma by Box-Behnken method,and provide theoretical basis for industrialized production of Gastrodiae Rhizoma.Methods Box-Behnken response surface method was used to optimize the freeze-drying process of Gastrodiae Rhizoma by taking the comprehensive scores of extract,gastrodin,p-hydroxybenzyl alcohol and barisenoside contents as indicators to determine the effect of pre-freezing time,drying temperature and primary drying time on the vacuum freeze drying process for Gastrodiae Rhizoma.Results The optimum freeze-drying process for Gastrodiae Rhizoma was as follows:the pre-freezing time was 10 h,the primary drying time was 18 h,and the drying temperature was 45℃.Conclusion The optimized lyophilization process for Gastrodiae Rhizoma is repeatable and feasiable,which can be used in the industrial production.
