马钱子治疗骨关节炎的潜在药物靶标

Nux vomica as a potential drug target for the treatment of osteoarthritis

目的探讨马钱子能否作为治疗骨关节炎(OA)的潜在药物靶标。方法在中药系统药理学技术平台(TCMSP)中根据口服生物利用度(OB)和类药性(DL)筛选出马钱子的化学成分及其相对应的靶基因;检索GeneCards、OMIM等数据库筛选OA疾病的相关靶点,应用Cytoscape3.7.2软件构建药物-活性成分-基因-疾病的网络,最后通过string平台构建蛋白相互作用网络,并筛选关键蛋白;同时对靶点基因作GO功能和KEGG通路富集分析。结果根据OB和DL筛选出13个马钱子主要活性成分,与马钱子治疗OA相关的靶基因17个;并筛选出10个关键蛋白和5条信号通路,而这些靶点和通路主要与镇痛、抗炎、核受体以及雌激素信号通路相关。结论马钱子治疗OA具有多途径、多成分、多靶点特点,核受体家族可能是马钱子治疗骨关节炎潜在药物靶标。

Objective To investigate whether nux vomica can be used as a potential drug target for the treatment of osteoarthritis(OA).Methods Based on the oral bioavailability(OB)and drug like(DL),the chemical constituents and their corresponding target genes were screened out in TCMSP.Searching genecards,OMIM and other databases to screen the related targets of OA disease,the network of drug active component gene disease was constructed by using Cytoscape 3.7.2 software.Finally,the protein interaction network was constructed by string platform,and the key proteins were screened.At the same time,go function and KEGG pathway enrichment of target genes were analyzed.Results According to OB and DL,13 main active components of nux vomica were screened,and 17 target genes related to the treatment of OA were identified.Ten key proteins and five signaling pathways were screened,and these targets and pathways were mainly related to analgesic,anti-inflammatory,nuclear receptor and estrogen signaling pathways.Conclusion Nux vomica has the characteristics of multi-channel,multi-component and multi-target in the treatment of OA.The nuclear receptor family may be the potential drug target of nux vomica in the treatment of osteoarthritis.
