核桃楸提取物抑制人卵巢癌细胞HO-8910PM增殖的实验研究 被引量:1

摘要 目的研究中药核桃楸提取物对人卵巢癌细胞HO-8910PM增殖能力的影响。方法采用层流分离法提取出核桃楸6种有效成分,分别以10^-8 mg/mL、10^-7 mg/mL、10^-6 mg/mL、10^-5 mg/mL浓度作用于卵巢癌细胞HO-8910PM,并设立空白对照组,以噻唑蓝比色法和形态学观察法检测核桃楸提取物对癌细胞生长的抑制作用。结果核桃楸提取物的6种有效成分结构鉴定分别为a-羟基-四氢萘醌(Y1)、胡桃醌(Y2)、4,8-二羟基萘酚-1-0-β-D-[6′-0-(3″,5″-二甲氧基-4″-羟基苯甲酰基)]葡萄糖苷(Y3)、4,8-二羟基萘酚-β-D-(6′-乙酰氧基葡萄糖苷)(Y4)、4,8-二羟基萘酚-β-D-葡萄糖苷(Y5)、4,5,8-二羟基-ɑ-四氢萘醌-5-0-β-D-[6′-0-(4″-羟基-3″,5″-二甲氧基苯甲酰基)]葡萄糖苷(Y6)。Y1、Y3、Y6这3种药物随着药物浓度的增高和作用时间的延长,对HO-8910PM细胞增殖的抑制率也相应增高,并且Y1、Y3的浓度依赖性明显,Y6的时间依赖性明显且在短时间低浓度组(10^-8 mg/mL)表现出了明显的肿瘤抑制作用;其他药物无规律变化。电镜下观察各药物组HO-8910 PM细胞随着药物浓度的增高和作用时间的延长,细胞出现程序性死亡。结论核桃楸提取物的6种有效成分中,有3种能够抑制人卵巢癌细胞HO-8910PM的增殖,且抑制作用存在剂量、时间依赖性。 Objective It is to investigate the inhibitory effect of extractors from Juglans Mandshurica Maxim onproliferation of ovarian carcinoma cells HO-8910PM.Methods Laminar flow separation method was used to extract 6 kinds of effective components of Juglans Mandshurica Maxim,which were used to treated for ovarian cancer cell HO-8910PM at 10^-8 mg/mL,10^-7 mg/mL,10^-6 mg/mL,10^-5 mg/mL concentration respectively,and a blank control group was established,and the inhibitory effect of Juglans Mandshurica Maxim extract on the growth of cancer cells was detected by thiazole blue colorimetry and morphological observation.Results The molecular structure of six extractors from Juglans Mandshurica Maxim was authenticated as a-Hydroxy-tetrahydronaphthoquinone(Y1),juglone(Y2),4,8-dihydroxynaphthol-1-0-β-D-[6′-0-(3″,5″-dimethyl Oxy-4″-hydroxybenzoyl)]glucoside(Y3),4,8-dihydroxynaphthol-β-D-(6′-acetoxyglucoside)(Y4),4,8-di Hydroxynaphthol-β-D-glucoside(Y5),4,5,8-dihydroxy-ɑ-tetrahydronaphthoquinone-5-0-β-D-[6′-0-(4″-hydroxy-3″,5″-Dimethoxybenzoyl)]glucoside(Y6).With the increase of drug concentration and the prolongation of the action time of the three drugs Y1,Y3,and Y6,the inhibition rate of HO-8910PM cell proliferation also increased,and the concentration dependence of Y1,Y3 was obvious,and the time dependence of Y6 was obvious.And in the short-term low-concentration group(10^-8 mg/mL),it showed obvious tumor suppression;other drugs changed irregularly.Under the electron microscope,the HO-8910 PM cells in each drug group were observed to undergo programmed cell death as the drug concentration increased and the action time prolonged.Conclusion Among the six active ingredients of Juglans Mandshurica extract,three kinds can inhibit the proliferation of human ovarian cancer cells HO-8910PM,and the inhibitory effect is dose-and time-dependent.
