锌铁转运蛋白基因OsZIP5和OsZIP9参与水稻Zn^2+和Cd^2+的吸收和转运 被引量:4

摘要 锌(Zn)和镉(Cd)在水稻中的积累与世界粮食营养健康与安全息息相关。锌和镉具有相似的化学和物理性质,但在高等生物中具有不同的生理效应。在植物中,通过不同基因家族的不同转运蛋白参与锌和镉的吸收或转运。本研究对水稻锌铁转运蛋白(zinc-regulated transporters, iron-regulated transporter-like proteins,ZIPs)基因家族的16个成员进行生物信息学分析,并在锌缺乏或加镉胁迫条件下开展其表达模式分析,探究ZIP基因家族成员在水稻锌和镉吸收及累积过程中可能的生物学功能。研究发现,Os ZIP5和Os ZIP9同时受锌缺乏和镉胁迫的诱导表达。通过异源酵母表达验证了Os ZIP5和Os ZIP9具有Zn^2+和Cd^2+的转运活性,并进一步通过不同时期不同组织的表达模式分析发掘Os ZIP5和Os ZIP9在水稻Zn^2+和Cd^2+吸收或转运中的作用。本研究为水稻ZIP基因家族成员参与Zn2+和Cd2+的吸收和累积的相关研究提供参考。 The accumulation of zinc(Zn)and cadmium(Cd)in rice is closely related to the health and safety of world food nutrition.Zinc and cadmium have similar chemical and physical properties,but have different physiological effects in higher organisms.In plants,different transport proteins of different gene families are involved in the absorption and transport of zinc and cadmium.In this study,bioinformatics analysis was performed on 16 members of the rice ZIP(zinc-regulated transporters,iron-regulated transporter-like proteins,ZIPs)gene family,and the expression patterns of different members under zinc deficiency and cadmium stress conditions were analyzed to explore the possible biological functions of members of the ZIP gene family in the absorption and accumulation of zinc and cadmium in rice.It was found that Os ZIP5 and Os ZIP9 were simultaneously induced by zinc deficiency and cadmium stress.Through heterologous expression in yeast,it was verified that Os ZIP5 and Os ZIP9 have Zn^2+and Cd^2+transporting activities.Further,the role of Os ZIP5 and Os ZIP9 in the absorption or transport of Zn^2+and Cd2+in rice was analyzed by studying their expressions patterns in various tissues at different growth stages.This study provides a reference for related studies on the members of the rice ZIP gene family involved in the absorption and accumulation of Zn^2+and Cd^2+.
