基于TCGA数据库分析KIF20A在子宫内膜癌中的表达和意义

Expression of KIF20A in endometrial carcinoma and its clinical significance:a study by bioinformatics analysis

目的检测驱动蛋白家族成员20A(KIF20A)在子宫内膜癌中的表达及其预后价值。方法下载癌症基因组图谱(TCGA)中子宫内膜癌组织和癌旁组织样本的基因表达谱数据和临床数据,比较KIF20A在子宫内膜癌组织和癌旁组织中的表达,并检测KIF20A表达水平与患者生存预后的关系。结果子宫内膜癌组织中KIF20A表达高于癌旁组织(P<0.001)。KIF20A表达水平与病理类型、FIGO分期及组织学分级显著相关(P均<0.05)。Kaplan-Meier法显示,子宫内膜癌患者中,KIF20A高表达组患者的无病生存率显著低于低表达组(P=0.002);子宫内膜样腺癌中,KIF20A高表达组的无病生存率仍低于低表达组(P=0.003)。多因素Cox回归分析显示,KIF20A高表达和淋巴结阳性是影响子宫内膜样腺癌患者无病生存率的独立预后因素(P均<0.05)。结论KIF20A在子宫内膜癌组织中较癌旁正常组织高表达,KIF20A可能是子宫内膜样腺癌患者预后的独立影响因素。

Objective:To assess the kinesin family 20A(KIF20A)expression in endometrial carcinoma and its prognostic value.Methods:The RNA-seq expression data and clinical data in the cancer genome atlas(TCGA)were downloaded.The expression of KIF20A in endometrial carcinoma tissues and adjacent nonneoplastic tissues was compared.The relationship between the expression level of KIF20A and prognosis of patients was detected.Results:KIF20A expression level in endometrial carcinoma specimens was significantly increased(P<0.001).KIF20A expression was significantly related to pathological type,FIGO stage and histologic grade(P<0.05).Kaplan-Meier method showed that patients with tumors expressing high KIF20A levels had poorer disease-free survival than patients with tumors expressing low KIF20A levels(P=0.002).Similar trends of DFS in endometrioid endometrial carcinoma had also been observed(P=0.003).The Multivariate Cox proportional hazards model showed that KIF20A expression and lymph nodes positive were independent poor prognostic factor for disease-free survival(P<0.05)in endometrioid endometrial carcinoma.Conclusion:KIF20A is over-expressed in endometrial carcinoma.KIF20A maybe an independent marker of prognosis in endometrioid endometrial carcinoma.
