绿原酸治疗口腔溃疡的动物实验研究 被引量:4

摘要 目的探讨绿原酸对大鼠口腔溃疡的治疗效果,分析其对肿瘤坏死因子α(TNF-α)、超氧化物歧化酶(SOD)的影响。方法采用化学灼烧法在72只雌性SD大鼠左右两侧颊黏膜建立口腔溃疡模型。将大鼠随机分为四组:高浓度组(10.0mg/mL绿原酸溶液)、中浓度组(5.0mg/mL绿原酸溶液)、低浓度组(2.5mg/mL绿原酸溶液)和对照组。造模后,高、中、低浓度组溃疡表面局部给予绿原酸溶液,每天2次,对照组溃疡表面不予处理。各组分别于首次给药后第3、5、7天处死大鼠6只,比较大鼠口腔溃疡黏膜面愈合情况,并对溃疡组织进行病理学观察,测定血清中TNF-α、SOD的变化水平。结果高、中、低浓度组的溃疡面积与对照组比较,差异有统计学意义(P <0.05)。各浓度组的固有层炎症细胞明显减少,上皮修复明显。实验第3、5、7天,低、中、高浓度组的TNF-α活性均明显降低,血清SOD水平增加,与对照组比较差异有统计学意义(P <0.05)。结论绿原酸可促进大鼠口腔溃疡愈合,具有抗炎和抗氧化作用。 Objective To investigate the therapeutic effect of chlorogenic acid on oral ulcer in rats and analyze its effects on tumor necrosis factor-α(TNF-α) and superoxide dismutase(SOD). Methods A model of oral ulcer was established on the buccal mucosa on the left and right sides of 72 female SD rats by chemical burning. These rats were randomly divided into four groups: High concentration group(10 mg/mL chlorogenic acid solution), medium concentration group(5 mg/mL chlorogenic acid solution), low concentration group(2.5 mg/mL chlorogenic acid solution) and control group. After modeling, the ulcer surfaces of the high, medium and low concentration groups were locally treated with the corresponding chlorogenic acid solution twice a day, while the ulcer surface of the control group was not treated. Six rats in each group were sacrificed on days 3, 5 and 7 after the first administration, respectively. The healing of mucosal surface of the oral ulcer in rats was compared, the histopathological changes of ulcers were observed, and the serum levels of TNF-α and SOD were measured. Results The area of ulcers,in the high, medium, and low concentration groups was sigificantly different from that of the control group(P < 0.05). Lamina propria inflammatory cells were significantly reduced, and the epithelium was obviously repaired in each concentration group. On days 3, 5 and 7 of the experiment, the activity of TNF-α in the low, medium and high concentration groups was significantly decreased, and the serum level of SOD was increased, which was statistically significant compared with the control group(P < 0.05). Conclusion Chlorogenic acid can promote the healing of oral ulcer in rats and has anti-inflammatory and antioxidant effects.
