摘要
刺芹侧耳(Pleurotus eryngii)具有较强的木质纤维素降解能力,可以以多种农业废弃物为生产原料。为研究其在不同栽培原料上的生长性能及木质素降解酶类的诱导分泌情况,以刺芹侧耳52611为试验材料,观察该菌株在棉籽壳、木屑、玉米芯、玉米秸秆、甘蔗渣、花生壳和柠条7种农林废弃物基质上的菌丝生长速度,并测定其液体发酵胞外产漆酶、木质素过氧化物酶的情况。结果显示,以玉米芯、玉米秸秆、木屑、花生壳为基质的培养基上,菌丝长速和长势较好,显著优于棉籽壳、柠条和甘蔗渣基质上的菌丝生长状况,其中甘蔗渣培养基质上的菌丝长势最弱;液体发酵胞外酶活性的测定结果显示,在菌丝长速和长势较好的这些基质的液体发酵液中,漆酶和木质素过氧化物酶的活性也相应较高,说明菌丝的生长速度与诱导产生的木质纤维素酶类的活性紧密相关,活性越高,所能释放的碳源越多,菌丝生长状况就越好。研究结果为刺芹侧耳培养基质的优化及生产效益的提升提供重要科学依据。
Pleurotus eryngii has strong lignocellulose degradation ability and can use a variety of agricultural wastes as raw materials. In order to study its growth performance on different cultivation materials and the induced secretion of lignin-degrading enzymes, using P. eryngii 52611 as experimental material to measure the mycelial growth rate of the strain on cottonseed hull,sawdust, corncob, corn straw, bagasse, peanut shell and korshinsk peashrub branches, the activities of extracellular laccase and lignin peroxidase under liquid fermentation condition were determined. The results showed that the mycelial growth rate and vigor on the medium with corncob, corn straw, sawdust and peanut shell grew better, which was significantly better than that on cottonseed shell, korshinsk peashrub branches and bagasse substrates, the mycelial growth vigor on bagasse culture medium being the weakest. The results of extracellular enzyme activity in liquid fermentation showed that the activities of laccase and lignin peroxidase were also higher in the liquid fermentation broth of these substrates with mycelium growth rate and vigor being better, showing that the growth rate of mycelium was closely related to the activity of induced lignocellulase, the higher the activity, the more carbon sources could be released, and the better the mycelial growth was. The results can provide an important scientific basis for the optimization of culture medium and the improvement of production efficiency of P. eryngii.
