摘要
研究榼藤提取物对牛Ⅱ型胶原诱导的类风湿性关节炎(collagen-induced arthritis,CIA)大鼠模型的治疗作用.通过牛Ⅱ型胶原与弗氏佐剂来诱导大鼠CIA模型,建模期间通过体征观察和足趾肿胀度评分来判定建模是否成功.将造模成功的大鼠随机分为模型组,榼藤提取物低(93.75 mg/kg)、中(187.50 mg/kg)、高(375.00 mg/kg)剂量组,雷公藤多苷组和地塞米松组.结果表明榼藤提取物可以降低CIA大鼠足掌及踝关节肿胀度;降低脾脏指数;下调血清中IL-1β、IL-17、PGE2的含量;明显减轻滑膜增生,减少炎细胞浸润及血管翳的形成,并改善软骨病变损伤;体外细胞实验结果表明榼藤提取物可能是通过抑制IL-1β、TNF-α、PGE2等相关炎性因子的表达起到抗炎的作用.榼藤提取物能够减轻CIA大鼠类风湿关节炎的症状,可能与其发挥抗炎作用下调IL-1β、IL-17、TNF-α、PGE2等炎性因子表达有关.
To study the therapeutic effect of Entada phaseoloides extract(EP)on bovine typeⅡcollagen-induced rheumatoid arthritis(collagen-induced arthritis,CIA)model rats,rats CIA model was induced by bovine typeⅡcollagen and Freund's adjuvant(IFA).During the modeling period,whether the modeling was successful or not was determined by physical observation and toe swelling score.Immunized CIA rats were randomly divided into model group,EP-low(93.75 mg/kg)group,EP-medium(187.50 mg/kg)group,EP-high(375.00 mg/kg)group,Tripterygium wilfordii group and dexamethasone group.The results showed EP could reduce the degree of paw swelling of CIA rats,significantly alleviate synovial hyperplasia,mitigate infiltration of inflammatory cells and the formation of vasospasm,and improve cartilage lesions,lower the spleen index,as well as down-regulating the production of IL-1β,IL-17,PGE2 in serum.On the other hand,the results of cell experiments indicated that the extract of EP might play an anti-inflammatory role by inhibiting the expression of IL-1β,TNF-α,PGE2 and other related inflammatory factors in vitro.The Entada phaseoloides extract could relieve the symptoms of rheumatoid arthritis in CIA rats,which may be related to the effect of down-regulation IL-1β,IL-17,TNF-αand PGE2 production..
