摘要
目的检测胃癌组织内差异表达的长链非编码RNA(long non-coding RNA,lncRNA)的表达情况,探寻新的胃癌标志物。方法选取5对胃癌患者的癌组织及其配对癌旁正常胃黏膜组织标本,应用人类lncRNA芯片进行筛查,分析差异表达的lncRNA谱,综合评判选定目标lncRNA,再扩大样本量行实时荧光定量聚合酶链反应检测,分析其表达量与胃癌临床参数的相关性。结果芯片筛查发现5445个差异表达的lncRNA。lncRNA p2819在胃癌组织中的相对表达量显著低于癌旁正常组织(P<0.05),且lncRNA p2819的相对表达量与胃癌分化程度、TNM分期均呈负相关(均P<0.05),而与性别、年龄、肿瘤位置、肿瘤最大直径等均无显著相关性(均P>0.05)。结论胃癌组织中lncRNA p2819表达下调,其表达量与胃癌恶性程度、TNM分期呈显著负相关,可作为胃癌的潜在标志物。
Objective To investigate long non-coding RNA(lncRNA)expression profile in gastric cancer tissue and to identify new biomarker for gastric cancer.Method The expression profile of lncRNA were detected in 5 paired gastric cancer and adjacent normal tissues by microarray.The target lncRNA was selected by comprehensive analysis and judgement.The expression was reassessed by quantitative reverse transcription-polymerase chain reaction(qRT-PCR)test in an increased sample volume.The association between the expression of target lncRNA and clinical features were analyzed.Result Five thousand four hundred and forty five differentially expressed lncRNAs were found by microarray.The relative expression of lncRNA p2819 was significantly down regulated in gastric cancer tissues than those in paired adjacent tissues(P<0.05).The relative expression of lncRNA p2819 is negatively correlated with the malignancy of gastric cancer and its TNM staging(all P<0.05),but has no significant correlation with gender,age,tumor location and tumor maximum diameter(all P>0.05).Conclusion The expression of lncRNA p2819 was down regulated in gastric cancer tissue,and its expression is negatively correlated with the malignancy of gastric cancer and TNM staging.LncRNA p2819 may serve as a potential new biomarker of gastric cancer.
