虾青素对过氧化氢诱导的骨髓间充质干细胞氧化应激的保护作用 被引量:3

Protective effect of astaxanthin on H2O2-induced oxidative stress in bone marrow mesenchymal stem cells

摘要 目的探讨虾青素对过氧化氢(H2O2)诱导的骨髓间充质干细胞(BMSC)氧化应激的保护作用及可能机制。方法体外培养人BMSC,采用H2O2建立氧化应激模型,应用不同浓度的虾青素预保护BMSC。CCK-8法检测细胞存活率,流式细胞术检测细胞凋亡率,生物化学法测定细胞中谷胱甘肽(GSH)含量、超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量。Western blotting检测细胞中核因子E2相关因子2(Nrf2)、血红素加氧酶-1(HO-1)、还原型辅酶/醌氧化还原酶1(NQO1)蛋白含量。结果虾青素明显提高H2O2诱导的氧化应激BMSC的存活率,提高细胞内SOD活性、GSH含量,降低细胞内MDA含量,降低细胞凋亡率,提高Nrf2蛋白表达量。结论虾青素可能通过激活Nrf2/抗氧化响应元件通路,减轻H2O2诱导的BMSC氧化应激。 Objective To investigate the effect of astaxanthin on hydrogen peroxide(H2O2)-induced oxidative stress response and the possible mechanism of that response in bone marrow mesenchymal stem cells(BMSCs).Methods Human BMSCs were cultured in vitro.H2O2 was used to induce oxidative stress in BMSCs.BMSCs were pretreated with varying concentrations of astaxanthin.Cell survival was analyzed via a CCK-8 assay,and cell apoptosis was detected using flow cytometry.A biochemical method was used to measure glutathione and malonyldialdehyde levels as well as superoxide dismutase activity.Western blotting was employed to determine nuclear factor erythroid-2-related factor 2(Nrf2),heme oxygenase 1,and quinone oxidoreductase 1 expression levels.Results Astaxanthin significantly improved the survival rate of BMSCs that had been subjected to H2O2-induced oxidative stress;it also increased superoxide dismutase activity,increased glutathione level,and reduced malondialdehyde level.Furthermore,astaxanthin lowered the cell apoptosis rate and induced Nrf2 protein expression.Conclusion Astaxanthin may attenuate H2O2-induced oxidative stress in BMSCs by activating the Nrf2/antioxidant response element pathway.
