水中产气荚膜梭状芽孢杆菌的检测 被引量:2

摘要 《生活饮用水卫生标准》(GB 5749-2006)附录A中对产气荚膜梭菌给出了0 CFU/(100 m L)的限量值,但并未指定相应的检测方法。因此,对目前现行有效的3个检测产气荚膜梭菌的检测标准(GB 4789.13-2012、GB/T 8538-2016和SN/T 0177-2011)进行汇总对比。对购买的产气荚膜梭菌菌种,使用胰胨-亚硫酸铵-环丝氨酸琼脂培养基(TSC)、液体硫乙醇酸盐培养基(FTG)和疱肉培养基,分别进行复苏培养,通过对各自同代传代产物开展不同的性能验证试验,确定其复苏效果。结果显示,这3种培养基对该菌菌种均有良好的复苏能力。产气荚膜梭菌的传统检测方法检测步骤多,涉及的培养基种类多,且检测时间长。基于此,对比传统检测方法,使用显色培养基,对南方某自来水公司的出厂水、水源水和南方某污水厂进水、二沉池出水中的产气荚膜梭菌含量进行检测。结果显示,显色培养基与传统检测方法的检测结果一致,但所耗费的检测时间和所需的培养基种类却明显优于传统检测方法。 Limit value of 0 CFU/(100 mL)for Clostridium perfringens is given in Appendix A of hygienic standard for drinking water(GB 5749—2006),but the corresponding detection method is not specified.Therefore,author summarized and compared three currently effective detection standards for Clostridium perfringens(GB 4789.13—2012,GB/T 8538—2016 and SN/T 0177-2011).Purchased Clostridium perfringens strains were resuscitated with tryptone ammonium sulfite cycloserine agar medium(TSC),liquid thioglycolate medium(FTG)and cooked meat medium,respectively.Recovery effect was determined by carrying out different performance verification experiments on the same generation products.Results showed that the three kinds of media had good recovery ability for the strain.Traditional detection method of Clostridium perfringens consists of many steps,involves many kinds of culture medium,and necessitates a long detection time.Concentration of Clostridium perfringens in tap water,source water of a water supply company,influent,secondary sedimentation tank of a wastewater treatment plant in South China were detected by using chromogenic medium and traditional detection method respectively.It showed results of using chromogenic medium were closed to results of traditional detection methods,Detection time and the type of culture medium needed were significantly better than those of traditional detection methods.
