摘要
采用高效液相色谱-紫外检测器法检测大米中维生素B1的含量,并对色谱条件、样品中维生素B1的提取方法等进行优化。经优化后的条件:流动相为乙酸钠(0.05 mol/L)∶甲醇(体积比)=80∶20、柱温30℃、流动相流速1.0 mL/min,样品中加入20.0 mL浓度为0.01 mol/L的盐酸溶液进行溶解并超声提取45 min,同时进行了样品加标回收实验和方法最低检出限等实验,回收率92%以上,最低检出限0.17 ng/mL,具有操作简便、快速、准确等特点。
The content of vitamin B1 in rice was determined by HPLC-UV detector,the chromatographic conditions and extraction methods of vitamin B1 in samples were optimized. Optimized condition was: the mobile phase is sodium acetate (0. 05 mol/L)∶methanol (volume ratio) = 80∶20,column temperature 30 ℃,the flow rate of the mobile phase 1. 0 mL/min,20. 0 ml hydrochloric acid solution with a concentration of 0. 01 mol/L was added to the sample for dissolution and ultrasonic extraction for 45 min,at the same time,the experiments were carried out on the recovery of the sample and the minimum detection limit of the method,the recovery rate was above 92 %, and the minimum detection limit was 0. 17 ng/m L,which was characterized by simple,rapid and accurate operation.
