外积融合光谱结合HPLC快速测定白术内酯Ⅰ、Ⅱ、Ⅲ含量 被引量:1

摘要 目的采用外积融合光谱结合HPLC建立白术内酯Ⅰ、Ⅱ、Ⅲ的定量分析模型,以实现快速检测。方法收集不同产地白术饮片123批,采用HPLC测定白术内酯Ⅰ、Ⅱ、Ⅲ的含量,同时采集X射线荧光光谱和近红外光谱,采用多维偏最小二乘法建立3种白术内酯的定量分析模型。结果白术中白术内酯Ⅰ、Ⅱ、Ⅲ的外积融合光谱定量分析模型的决定系数(R~2)分别为0.946 4、0.917 2、0.955 8,校正均方根误差(RMSE_C)分别为0.021 8、0.018 1、0.040 0,预测均方根误差(RMSE_P)分别为0.031 1、0.025 8、0.049 8。结论本研究建立的白术内酯Ⅰ、Ⅱ、Ⅲ外积融合光谱定量分析模型具有较好的准确性,可用于白术内酯Ⅰ、Ⅱ、Ⅲ的快速测定,为快速评价白术饮片质量提供依据。 Objective To establish a rapid quantification method for atractylenolides in Astractylodis Macrocephalae Rhizoma by outer product fusion(OPF)spectra and HPLC;To realize the rapid determination.Methods Totally 123 batches of Astractylodis Macrocephalae Rhizoma decoction pieces from different origins were collected.The contents of atractylenolideⅠ,ⅡandⅢwere determined by HPLC.X-ray fluorescence spectroscopy and near infrared spectroscopy of samples were also collected.The quantitative analysis model of three atractylenolides was established by N-way partial least square method.Results The determination coefficients(R2)of atractylenolideⅠ,ⅡandⅢin OPF spectra analysis models were 0.9464,0.9172,0.9558;root mean square error of calibration(RMSEC)were 0.0218,0.0181,0.0400;root mean square error of prediction(RMSEP)were 0.0311,0.0258,0.0498,respectively.Conclusion The established OPF quantitative analysis model of three types of atractylenolide in Astractylodis Macrocephalae Rhizoma has a good accuracy,which can be used for the rapid determination of atractylenolideⅠ,Ⅱ,andⅢ,providing a basis for the rapid evaluation of the quality of Astractylodis Macrocephalae Rhizoma decoction pieces.
