茶轮斑病致病病原菌的鉴定 被引量:8

摘要 为茶轮斑病的防控提供理论依据,采用单孢分离法结合形态学与分子生物学鉴定方法研究茶轮斑病的致病病原菌。结果表明:从植株发病部位分离的菌株SQ-20-1形态学特征与棕榈拟盘多毛孢(Pestalotiopsis trachicarpicola)相似;菌株SQ-20-1回接致病症状与田间发病症状一致;经NCBI的BLAST比对,其DNA的ITS、tef1和β-tubulin序列与P.trachicarpicola的ITS、tef1及β-tubulin序列的同源性较高,分别达99.8%、100%和100%;联合ITS、tef1和β-tubulin的3个基因片段构建最大似然和最大简约系统发育树,菌株SQ-20-1与P.trachicarpicola处于同一分支,支持率达66/97。因此,结合形态学特征和分子生物学特性,确定引起茶轮斑病的病原菌为棕榈拟盘多毛孢。 In order to provide theoretical basis for prevention and control of tea gray blight,the pathogen of tea gray blight was studied by using single spore isolation combined with morphological and molecular biological identification methods.Results:The morphological characteristics of strain SQ-20-1 isolated from diseased plant were similar to Pestalotiopsis trachicarpicola;The pathogenetic symptoms of strain SQ-20-1 were consistent with those of field disease;By Blast at NCBI,the homology in ITS,tef1 andβ-tubulin sequences between DNA and P.trachicarpicola reached 99.8%,100%and 100%,respectively;The maximum likelihood and maximum parsimony phylogenetic tree was constructed by combining three gene segments of ITS,tef1 andβ-tubulin sequences,strain SQ-20-1 and P.trachicarpicola were in the same branch with 66/97 support rate.Therefore,according to morphological characteristics and molecular biological characteristics,it is determined that P.trachicarpicola is the pathogen causing tea leaf spot disease.
