原发性骨质疏松症伴中、重度骨痛患者iRhom2/TACE信号通路表达 被引量:1

Expression of iRhom2/TACE signaling pathway in patients with primary osteoporosis with moderate or severe bone pain

摘要 目的探讨原发性骨质疏松症(osteoporosis,OP)伴中、重度骨痛患者非活性菱形蛋白2(inactive rhomboid-like protein 2,iRhom2)/肿瘤坏死因子α转化酶(tumor necrosis factor-αconverting enzyme,TACE)信号通路表达变化。方法107例原发性OP患者按照是否伴中、重度骨痛(VAS均≥4分)分为OP伴骨痛组(57例)及OP组(50例),另外纳入50例健康体检者作为正常对照组。分别记录3组一般情况及VAS评分,Western blot法检测3组血清iRhom2、TACE及TACE前体蛋白(pro-TACE)表达水平,与VAS评分行相关性分析。结果OP伴骨痛组患者血清iRhom2、TACE表达水平明显高于OP组及正常对照组(P<0.05),而OP组及正常对照组血清iRhom2、TACE表达水平差异无统计学意义(P>0.05)。3组血清pro-TACE表达水平差异无统计学意义(P>0.05)。受试者血清iRhom2及TACE蛋白表达水平与疼痛VAS评分程度呈明显正相关性(r=0.885,r=0.914,P<0.05),与OP患者骨密度测定的T值无明显相关性(r=0.315,r=0.442,P>0.05)。结论iRhom2/TACE信号通路异常表达可能参与了骨质疏松性中、重度骨痛的病理过程,但患者骨密度值与iRhom2/TACE信号通路异常无明显相关性。 Objective To investigate the expression of inactive rhomboid like protein 2(iRhom2)/TNF-αconverting enzyme(TACE)signal pathway in primary osteoporosis(OP)patients with moderate or severe bone pain.Methods 107 patients with primary OP were divided into OP with bone pain group(57 cases)and OP group(50 cases)according to whether they were accompanied with moderate or severe bone pain(VAS≥4 points),and 50 healthy people were included as normal control group.The general condition and VAS score of the patients were recorded.The expression levels of iRhom2,TACE,and pro TACE precursor protein(pro TACE)in the 3 groups were detected by Western blot,and the correlation with VAS score was analyzed.Results The expression levels of iRhom2 and TACE in OP patients with bone pain were significantly higher than those in OP group and healthy control group(P<0.05),but there was no significant difference between OP group and healthy control group(P>0.05).There was no significant difference in serum Pro TACE expression among the 3 groups(P>0.05).There was a significant positive correlation between irhom2,TACE protein expression and VAS score(r=0.885,r=0.914,P<0.05),but not with T value level of BMD with osteoporosis patients(r=0.315,r=0.442,P>0.05).Conclusion Abnormal expression of iRhom2/TACE signaling pathway may be involved in the pathological process of moderate or severe osteoporotic bone pain,but there is no significant correlation between the degree of BMD level and abnormal iRhom2/TACE signaling pathway.
