摘要
建立DPPH-HPLC方法快速筛选、鉴别和测定食凉茶中抗氧化活性成分。食凉茶提取物与80μg/mL DPPH自由基室温避光孵育30min后,13000r/min,离心10min,取上清进行高效液相色谱分析,以峰强度减少大于80%的成分,认定为潜在的抗氧化活性物质,结合对照品对照,确定抗氧化活性成分为东莨菪内酯、槲皮素、山奈酚。建立HPLC法同时测定3个抗氧化活性成分的含量,并评价其抗氧化能力。利用DPPH-HPLC方法可快速、灵敏、准确筛选食凉茶中的抗氧化活性成分,并对其进行测定和评价,为开发利用食凉茶中抗氧化活性成分提供科学依据。
After reaction with 80μg/mL DPPH radical for 30 minutes at room temperature,the extract was centrifuged at 13000r/min for 10 minutes,and the supernatant was analyzed by HPLC.The components with peak intensity reduced by more than 80%were identified as potential antioxidants.By comparison with reference materials,it was determined that the antioxidants were scopoletin,quercetin,and kaempferol.It was able to simultaneously determine the contents of three antioxidant active ingredients and evaluate their antioxidant capacity by HPLC.The DPPH-HPLC method is rapid,sensitive and accurate,which provides a scientific basis for the development and utilization of antioxidant active ingredients in shiliang tea.
