摘要
目的研究芝芪康艾颗粒(芝康颗粒)对免疫抑制小鼠免疫功能的调节作用。方法将174只清洁级昆明小鼠分为空白对照组、模型组、贞芪扶正颗粒组(阳性药组)和芝康颗粒3.3、6.6和13.2 g/kg组,每组29只。以80 mg/kg(ip)环磷酰胺建立免疫抑制小鼠模型,各组实验小鼠连续给药(ig)14 d后,检测小鼠脏器指数、吞噬细胞吞噬能力、血常规、T淋巴细胞亚群比例以及血清细胞因子IL-6、TNF-α、IFN-γ水平,并观察胸腺、脾等免疫器官组织病理变化。结果与空白对照组相比,模型组小鼠胸腺指数降低(P<0.01),碳廓清指数k与吞噬指数α降低,胸腺和脾显微结构萎缩、紊乱,外周血白细胞、淋巴细胞、红细胞数目以及血红蛋白含量降低(P<0.05,P<0.01,P<0.01,P<0.01);外周血CD8+细胞及血清中IL-6、TNF-α、IFN-γ含量下降(均P<0.01),CD4+/CD8+比值升高(P<0.01),表明造模成功。与模型组相比,贞芪扶正颗粒组小鼠胸腺指数、外周血白细胞与淋巴细胞数目升高(均P<0.05),胸腺和脾组织结构逐渐恢复,廓清指数k和吞噬指数α升高(均P<0.01);CD4+/CD8+比值降低(P<0.01)。芝康颗粒6.6 g/kg组外周血白细胞和淋巴细胞数目升高(均P<0.05);芝康颗粒13.2 g/kg组小鼠的胸腺指数恢复正常,胸腺和脾组织结构逐渐恢复,廓清指数k和吞噬指数α升高(P<0.01);芝康颗粒6.6、13.2 g/kg组CD8+百分数、IL-6、IFN-γ升高(P<0.01,P<0.01,P<0.05),CD4+/CD8+比值降低(P<0.01);芝康颗粒13.2 g/kg组血清中IL-6、TNF-α、IFN-γ含量升高(均P<0.01)。结论芝康颗粒可通过调节机体非特异性、特异性免疫反应,改善机体免疫状态,从而增强机体免疫功能。
ObjectiveTo investigate the immunoregulatory effect of Zhiqi Kang’ai granules on immune function in immunosuppressive mice.MethodsA total of 174 clean-grade Kunming mice were divided into 6 groups:the control group,model group,Zhenqi Fuzheng granule group(positive drug group),and Zhiqi Kang’ai granule 3.3,6.6 and 13.2 g/kg dose groups,with 29 mice in each group. The immunosuppressive model was established by intraperitoneal injection(ip)of cyclophosphamide at 80 mg/kg. After ig for 14 days,the organ index,phagocytic phagocytosis,blood routine index,T lymphocyte subsets,and the serum cytokine interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),interferon-γ(IFN-γ)level were detected,and the histopathological changes of immune organs such as thymus and spleen in mice were observed.Results Compared with the control group,the thymus index in model group was decreased(P<0.01),with the carbon clearance index k and the phagocytic index α in decline and the microstructures of the thymus and spleen atrophied in disorder. The number of peripheral blood leukocytes,lymphocytes,red blood cells,and the content of hemoglobin were declined(P<0.05,P<0.01,P<0.01 and P<0.01),with the CD8+cells in peripheral blood,the content of IL-6,TNF-α,IFN-γ declined(P<0.01),and CD4+/CD8+increased(P<0.01). These data indicated that the immunosuppressive model was created successfully. Compared with the model group,the thymus index,number of leukocytes and lymphocytes in peripheral blood in the Zhenqi Fuzheng granule group were increased(P<0.05)and the thymus and spleen tissue structures were gradually recovered,with the clearance index k and the phagocytic index α on the rise(P<0.01);the ratio of CD4+/CD8+was decreased(P<0.01). The number of white blood cells and lymphocytes in the Zhiqi Kang’ai 6.6 g/kg group were increased(P<0.05);the thymus index in the 13.2 g/kg Zhiqi kang’ai granule group of Zhiqi kang’ai granules returned to normal,the thymus and spleen tissue structures were gradually recovered,with the clearance index k and the phagocytic index α on the rise(P<0.01);while in the 6.6 and 13.2 g/kg groups,the levels of CD8+cells,IL-6 and IFN-γ were increased(P<0.01,P<0.01 and P<0.05),the ratio of CD4+/CD8+was decreased(P<0.01);in the 13.2 g/kg Zhiqi kang’ai granule group,the levels of IL-6,TNF-α and IFN-γ were increased(P<0.01).Conclusion The Zhiqi Kang’ai granules can improve the non-specific and the specific immunity in immunosuppressed mice by regulating their immune-related indicators,thereby enhancing the immune function of the body.
