富血小板血浆对大鼠宫颈糜烂的治疗作用及其对炎性因子表达的影响 被引量:5

摘要 目的探讨富血小板血浆(PRP)对大鼠宫颈糜烂的治疗作用及其对白细胞介素(IL)-1β、肿瘤坏死因子α(TNF-α)和IL-6表达的影响。方法将60只雌性SD大鼠分为对照组、假手术组、模型组和PRP组,采用苯酚胶浆制作大鼠宫颈糜烂模型;实验期间观察各组大鼠的行为学变化;实验结束后进行取材,称量宫颈组织质量并计算宫颈脏器指数;苏木素-伊红(HE)染色观察各组大鼠宫颈组织病理变化;ELISA检测各组大鼠血清IL-1β、TNF-α及IL-6水平;Western blot检测各组大鼠宫颈组织IL-1β、TNF-α、IL-6表达水平。结果PRP使大鼠的精神状态明显好转,食欲有所增加,阴道分泌物减少,红肿程度减轻。HE染色显示,PRP使大鼠宫颈组织病理变化减轻,上皮细胞糜烂现象得到改善。与对照组和假手术组相比,模型组大鼠宫颈质量和宫颈脏器指数明显增加(P<0.01),血清IL-1β、TNF-α、IL-6水平明显上升(P<0.01),宫颈组织IL-1β、TNF-α、IL-6表达水平明显上升(P<0.01)。与模型组相比,PRP组大鼠宫颈质量和宫颈脏器指数明显下降(P<0.05),血清IL-1β、TNF-α、IL-6水平明显下降(P<0.05),宫颈组织IL-1β、TNF-α、IL-6表达水平明显下降(P<0.05)。结论PRP可能通过下调炎性因子的表达使宫颈糜烂得到缓解。 Objective To explore the therapeutic effect of platelet-rich plasma(PRP)on cervical erosion in rats and its effect on the expressions of interleukin(IL)-1β,tumor necrosis factorα(TNF-α)and IL-6.Methods A total of 60 female SD rats were divided into the control group,the sham group,the model group and the PRP group.Rat cervical erosion model was made by phenol glue.The behavioral changes of rats in each group were observed during the experiment.After experiment,materials were taken,the cervical tissue mass was weighed and the cervical organ index was calculated.Hematoxylin-eosin(HE)staining was used to observe the pathological changes of rat cervical tissues in each group.ELISA was used to detect the levels of serum IL-1β,TNF-αand IL-6,and Western blot was used to detect the expression levels of IL-1β,TNF-αand IL-6 in the cervical tissues.Results PRP significantly improved the mental state of rats,increased appetite,decreased vaginal secretions,and reduced redness and swelling.HE staining showed that PRP reduced the pathological changes of rat cervical tissue and improved the erosion of epithelial cells.Compared with the control group and the sham group,the cervical mass and cervical organ index in the model group increased significantly(P<0.01),and IL-1β,TNF-α,and IL-6 levels in serum(P<0.01)and cervical tissues(P<0.01)increased significantly.Compared with the model group,the cervical mass and cervical organ index of rats in the PRP group significantly decreased(P<0.05),IL-1β,TNF-α,and IL-6 levels in serum(P<0.05)and cervical tissue(P<0.05)increased significantly.Conclusion PRP may relieve cervical erosion by down-regulating the expression of inflammatory factors.
