摘要
目的研究大蒜提取物对变应性鼻炎(AR)模型大鼠的治疗作用。方法用卵清蛋白(OVA)致敏,氢氧化铝为佐剂诱发建立AR大鼠模型。鼻炎康片作为阳性对照药物。将72只SD大鼠随机分为空白组、模型组、阳性对照组及大蒜提取物低、中、高剂量组。除空白组外,其余5组大鼠均进行AR造模,造模成功后灌胃给药;阳性对照组给予鼻炎康片466 mg·kg^-1;大蒜提取物低、中、高剂量组分别给予大蒜提取物(以蒜氨酸计)3.15,6.30,12.60 mg·kg^-1;空白组及模型组给予等量生理盐水。隔日进行行为学评分。连续给药10 d后,检测大鼠血清免疫球蛋白E(IgE)、嗜酸性粒细胞阳离子蛋白(ECP)、白细胞介素-5(IL-5)、白细胞介素-13(IL-13)和一氧化氮(NO)水平,用超高效液相色谱-串联质谱(UPLC-MS/MS)法测定大鼠血清、心、肝、脾、肺、肾和脑组织的硫化氢水平,并用苏木精-伊红(HE)染色观察鼻黏膜病理学组织。结果大蒜提取物能明显改善AR大鼠鼻黏膜病变程度,减轻AR症状,降低血清中IgE、ECP、IL-5、IL-13和NO水平,大蒜提取物高剂量组肺、脑组织中硫化氢水平均明显高于空白组(P<0.05)。结论大蒜提取物对AR大鼠有明显的治疗作用,大蒜含硫化合物有望用于开发硫化氢供体药物以治疗AR。
Objective To study the treatment effect of garlic extract on allergic rhinitis(AR)in rats.Methods Rats were sensitized by ovalbumin(OVA)combined with aluminum hydroxide as adjuvant to make the AR model.Biyankang Tablets were used as positive control drug.72 SD rats were randomly divided into blank group,model group,positive control group and garlic extract low,middle and high dose groups.Except for the blank group,the AR model was made in the other 5 groups,and the rats were admi-nistrated intragastrically after the model was successfully built.Biyankang Tablets with the dose of 466 mg·kg^-1 were given in the positive control group,and garlic extract(calculated as alliin)with the dose of 3.15,6.30 and 12.60 mg·kg^-1 was given in low,middle and high dose groups.The blank group and model group were given the normal saline.The rats were intragastrically administrated for 10 d,and the AR symptom scores were evaluated every other day.Then the levels of specific immunoglobulin E(IgE),eosinophil cationic protein(ECP),interleukin-5(IL-5),interleukin-13(IL-13)and NO in serum were detected.The hydrogen sulphide levels in serum,heart,liver,spleen,lung,kidney and brain of rats were determined by UPLC-MS/MS method.Pathological examination of nasal mucosa was performed by using hematoxylin-eosin(HE)staining.Results Garlic extract could significantly decrease the pathological changes of nasal mucosa in AR rats,and reduce the AR symptoms,and lower the levels of IgE,ECP,IL-5,IL-13 and NO in serum.The hydrogen sulphide levels in lung and brain of the garlic extract high dose group were significantly higher than those of the blank group(P<0.05).Conclusion Garlic extract has a good treatment effect on AR rats.Garlic sulfur compounds can be used to develop hydrogen sulphide donor drugs for the treatment of AR.
