摘要
目的:研究MiR-181b-5p在胃癌中的表达水平,并进行生物信息学分析,探讨其在肿瘤发生、发展中的作用。方法:在GEO数据库检索并下载胃癌的miRNA数据,提取数据进行独立样本T检验和绘制ROC曲线,进行Meta分析,预测MiR-181b-5p的靶基因,并进行GO功能和KEGG通路分析。结果:检索筛选后得到5个miRNA芯片数据集,Meta分析显示胃癌和正常胃组织的miR-181b-5p的表达水平有差异,差异有统计学意义(P=0.008)。预测得到145个miR-181b-5p的靶基因,GO功能分析表明靶基因主要富集在转录的调控、细胞增殖和细胞周期的调节、蛋白质结合的调节等方面,差异有统计学意义(P<0.05);KEGG通路分析显示其主要涉及背腹轴形成、肿瘤miRNA、雌激素信号通路、神经营养蛋白信号通路、肿瘤的中心碳代谢等,差异有统计学意义(P<0.05)。结论:miR-181b-5p的上调与胃癌的有密切的联系,对胃癌的诊断可能有重要的意义。
Objective To investigate the expression level of miR-181b-5p in gastric cancer,and to perform bioinformatics analysis and its role in tumorigenesis.Method Methods:The miRNA data of gastric cancer were downloaded from the GEO database,and the data were extracted for independent sample t-test,ROC curve drawing and meta-analysis.The bioinformatics analysis of miR-181b-5p involved target prediction analysis,gene ontology enrichment and KEGG pathway enrichment.Results Five miRNA microarray data were collected,and statistical analysis showed that mir-181b-5p had diagnostic value for gastric cancer.The meta-analysis showed that there were statistically significant differences in the expression levels of mir-181b-5p in gastric cancer and normal gastric tissues(P=0.008).145 target genes of mir-181b-5p were predicted,and the GO function analysis showed that the target genes were mainly concentrated in the regulation of transcription,cell proliferation,the regulation of cell cycle arrest,and regulation of protein binding(P<0.05).KEGG pathway analysis showed that it was mainly involved in Dorso-ventral axis formation,MicroRNAs in cancer,estrogen signaling pathway,neurotrophic protein signaling pathway,Fox O signaling pathway,MAPK signaling pathway,and central carbon metabolism in cancer(P<0.05).Conclusion MiR-181b-5p expression is significantly higher in gastric cancer,and it has certain diagnostic value for gastric cancer.
