血清微小RNA-424联合血清人胎球蛋白A抗体、精子相关抗原9抗体诊断乳腺癌的价值 被引量:1

摘要 目的探讨血清微小RNA-424(miRNA-424)联合血清人胎球蛋白A(AHSG)抗体、精子相关抗原9(SPAG9)抗体诊断乳腺癌的价值。方法将58例乳腺癌患者作为乳腺癌组,55例乳腺纤维腺瘤患者为良性疾病组,51例健康女性为健康对照组。采用实时荧光定量PCR检测各组血清miRNA-424相对表达水平,采用酶联免疫吸附试验检测各组血清AHSG抗体、SPAG9抗体水平。比较三组受试者血清miRNA-424相对表达水平、AHSG抗体、SPAG9抗体水平,分析三者与乳腺癌临床分期的关系,以及三者单独及联合检测诊断乳腺癌的效能。结果(1)健康对照组、良性疾病组、乳腺癌组的血清miRNA-424相对表达水平依次降低,血清AHSG抗体、SPAG9抗体水平依次升高(均P<0.05)。(2)乳腺癌Ⅰ期、Ⅱ期、Ⅲ期患者的血清miRNA-424相对表达水平依次降低,血清AHSG抗体、SPAG9抗体水平依次升高(均P<0.05)。(3)血清miRNA-424、AHSG抗体、SPAG9抗体水平三者联合检测诊断乳腺癌的受试者工作特征曲线下面积为0.927,高于血清miRNA-424水平单独检测(P<0.05),且三者联合诊断的准确度高于AHSG抗体单独检测(P<0.05)。结论乳腺癌患者血清miRNA-424相对表达水平下降、AHSG抗体和SPAG9抗体水平升高,且临床分期越高者miRNA-424相对表达水平越低、AHSG抗体和SPAG9抗体水平越高,三者联合检测诊断乳腺癌的效能较高,或可用于乳腺癌的早期诊断。 Objective To investigate the diagnostic value of serum microRNA-424(miRNA-424)combined with serum alpha2-Heremans-Schmid glycoprotein(AHSG)antibody and sperm-associated antigen 9(SPAG9)antibody for breast cancer.Methods Fifty-eight patients with breast cancer were selected as breast cancer group,and 55 patients with breast fibroadenoma as benign disease group,and 51 healthy women as healthy control group.Real-time fluorescence quantitative PCR(qRT-PCR)was used to detect the relative expression level of serum miRNA-424 in each group,enzyme-linked immunosorbent assay was used to detect the levels of serum AHSG antibody and SPAG9 antibody in each group.The relative expression level of serum miRNA-424,AHSG antibody level,and SPAG9 antibody level of the subjects were compared among the three groups,the relationships of the three indexes with clinical stage of breast cancer were analyzed,and the diagnostic efficiency of the three indexes alone or in combination detection for breast cancer was analyzed as well.Results(1)The relative expression level of serum miRMA-424 decreased and serum AHSG antibody and SPAG9 antibody levels increased in the sequence of the healthy control group,the benign disease group and the breast cancer group(all P<0.05).(2)The relative expression level of serum miRMA-424 decreased and serum AHSG antibody and SPAG9 antibody levels increased in the order of patients with stageⅠ,ⅡandⅢbreast cancer(all P<0.05).(3)The area under receiver operating characteristic curve of the combined detection of serum miRNA-424,AHSG antibody and SPAG9 antibody levels for diagnosing breast cancer was 0.927,which was higher than that of serum miRNA-424 level detection alone(P<0.05),and the diagnostic accuracy of the combined detection of the three indexes was higher than single AHSG antibody detection(P<0.05).Conclusion Patients with breast cancer experience a decline in the relative expression level of serum miRNA-424 and an increase in the levels of AHSG antibody and SPAG9 antibody,moreover,those with higher clinical stage have lower relative expression level of miRNA-424 and higher AHSG antibody and SPAG9 antibody levels,the combined detection of the three indexes achieves higher diagnostic efficiency for breast cancer,and may be applied to early diagnosis of breast cancer.
