痰喘宁合剂平喘、镇咳、祛痰作用研究

Anti-asthmatic,antitussive and expectorant effects of the Tanchuanning mixture

目的研究痰喘宁合剂平喘、镇咳、祛痰作用。方法采用卵蛋白所致大鼠哮喘模型,观察大鼠气道阻力(RL)的变化,运用ELISA法检测分析各组大鼠血清中Ig E、IL-4、IL-17和TNF-α水平,并在镜下进行实验大鼠肺泡灌洗液(bronchoalveolar lavage fluid,BALF) WBC及EOS计数,HE染色观察肺组织病理变化并对其评分。采用氯化乙酰甲胆碱及磷酸组胺引喘豚鼠,观察痰喘宁合剂的平喘作用。采用氨水引咳小鼠、枸橼酸引咳豚鼠,观察痰喘宁合剂的镇咳作用。采用小鼠气管段酚红排泌量实验,观察痰喘宁合剂的祛痰作用。结果痰喘宁合剂可降低哮喘大鼠气道阻力,降低血清IL-4、IL-17和TNF-α水平,减少BALF中WBC及EOS数量,改善肺组织损伤情况,延长豚鼠引喘潜伏期(P <0.05或P <0.01);延长豚鼠及小鼠咳嗽潜伏期,减少咳嗽次数(P <0.05或P <0.01);增加酚红排泌量(P <0.05或P <0.01)。结论痰喘宁合剂具有平喘、镇咳、祛痰作用。

Objective To investigate the anti-asthmatic,anti-tussive,and expectorant effects of Tanchuanning mixture.Methods An Asthma model in rats was established by sensitization and challenge with ovalbumin(OVA)and then airway resistance(RL)was observed.Serum levels of Ig E,IL-4,IL-17,and TNF-αwere analyzed by ELISAs.The numbers of WBCs and EOSs in BALF of rats were counted and HE staining of lung tissue was performed to observe pathological changes.Methacholine chloride and histamine phosphate were used to induce asthma in guinea pigs to observe the anti-asthmatic effect of Tanchuanning mixture.The anti-tussive effect of Tanchuanning mixture was observed in a cough mouse model induced by ammonia and guinea pig model induced by citric acid.A phenol red excretion test in the tracheas of mice was used to observe the expectorant effect of Tanchuanning mixture.Results Tanchuanning mixture reduced airway resistance,decreased IgE,IL-4,IL-17,TNF-αlevels in serum and the number of WBCs and EOSs in BALF,and alleviated lung tissue damage in asthmatic rats(P<0.05 or P<0.01).Tanchuanning mixture prolonged asthma and cough latencies in guinea pigs and mice,respectively,reduced the cough times in mice,and increased phenol red excretion in mice(P<0.05 or P<0.01).Conclusions Tanchuanning mixture has obvious anti-asthmatic,anti-tussive,and expectorant effects.
