摘要 目的研究转录因子21(TCF21)在卵巢癌细胞株中的表达及其对卵巢癌细胞株HEY恶性生物学行为的影响。方法通过实时荧光定量聚合酶链反应(qRT-PCR)和免疫组织化学检测TCF21在正常卵巢组织、卵巢癌细胞株和高级别浆液性卵巢癌组织中的差异表达,采用qRT-PCR检测药物性去甲基化后TCF21的表达水平。在低表达TCF21的HEY卵巢癌细胞株中过表达TCF21,通过平板克隆试验检测细胞的增殖,建立裸鼠皮下种植瘤模型,观察裸鼠体内成瘤能力。通过Transwell试验检测过表达TCF21后对卵巢癌细胞HEY迁移、侵袭能力的影响。结果TCF21甲基化使其在卵巢癌细胞中的蛋白及mRNA表达明显低于正常卵巢细胞和组织,外源性过表达TCF21可抑制HEY细胞克隆形成率和成瘤能力,明显降低肿瘤细胞的迁移、侵袭能力。结论TCF21过表达可抑制卵巢癌细胞恶性生物学行为。 Objective To investigate the expression of transcription factor 21(TCF21)in ovarian cancer cell lines and its effect on the malignant biological behavior of ovarian cancer cell line HEY.Methods The expression of TCF21 were examined by real time fluorescent quantitative polymerase chain reaction(qRT-PCR)and immunohistochemistry in normal ovarian tissue and ovariom cancer cell lines and high-grade serous ovarian cancer tissue.TCF21 expression after drug demethylation was detected by qRT-PCR.TCF21 was overexpressed in hey ovarian cancer cell line with low expression of TCF21.The proliferation of cells was detected by plate cloning experiment,and the subcutaneous tumor model was established in nude mice to observe the tumorigenicity in vivo.The effect of overexpression of TCF21 on the migration and invasion of ovarian cancer cells HEY was detected by Transwell chamber cell migration assay.Results The protein and mRNA expression of TCF21 methylation in ovarian cancer cells was significantly lower than that in normal ovarian cells and tissues.Exogenous overexpression of TCF21 could inhibit the clone formation rate and tumorigenicity of HEY cells,and significantly reduce the migration and invasion ability of tumor cells.Conclusion Overexpression of TCF21 inhibits the malignant biological behavior of ovarian cancer cells.
