DNA依赖蛋白激酶在自噬中的作用

摘要 DNA依赖性蛋白激酶(DNA-dependent protein kinase,DNA-PK)是一种丝氨酸/苏氨酸激酶,参与细胞内许多生物学过程,在非同源末端连接(Non-Homologous End-Joining,NHEJ)途径中的DNA双链断裂修复中起着重要作用。自噬是40多年前发现的,现今已知有3种主要类型的自噬:巨自噬、分子伴侣介导的自噬及微自噬。最近有研究发现DNA-PK在这3种自噬中通过NHEJ对DNA损伤进行修复来影响细胞中的自噬,还发现通过磷酸化热休克蛋白HSPA8/HSC70来选择性诱导自噬,本文就从DNA-PK在自噬中的联系与作用做一综述。 DNA-PK is a serine/threonine kinase,which is involved in many cellular biological processes and plays an important role in the repair of DNA double strand breaks in the non homologous end joining(NHEJ)pathway.Autophagy was discovered more than 40 years ago,Nowadays,there are three main types of autophagy:megaautophagy,chaperone autophagy and microautophagy.Recently,it has been found that DNA-PK can repair DNA damage in these three autophagies by NHEJ,and selectively induce autophagy by phosphorylation of hspa8/Hsc70.This article reviews the relationship and role of DNA-PK in autophagy.
