Ultrasonic Assisted Ionic Liquid Extraction of Total Flavonoids from Pomelo Peel and Their Antioxidant Activities

摘要 目的:优化超声辅助离子液体提取柚子皮总黄酮的工艺条件,并评价其体外抗氧化活性。方法:在单因素试验基础上,以料液比、超声功率、超声时间为影响因素,总黄酮得率为评价指标,正交试验设计法优化提取工艺。再通过比色法测定总黄酮对羟自由基、超氧阴离子清除的能力。结果:最佳提取参数条件为料液比1∶20,超声功率200 W,超声时间50 min,总黄酮得率为1.16%。清除率与总黄酮质量浓度存在明显的剂量关系,2.5μg/m L总黄酮对羟自由基、超氧阴离子的清除作用最强,清除率分别为43.8%、41.4%。结论:该方法稳定可行,重复性好,可用于离子液体提取有明显体外抗氧化活性的柚子皮总黄酮,为该体系的放大实验或规模化生产提供理论指导。 Objective:To optimize the ultrasonic extraction auxiliary ionic liquids pomelo peel process conditions of total flavonoids.The antioxidant activity was evaluated in vitro.Methods:On the basis of single factor experiment,taking the ratio of solid to liquid,ultrasonic power and ultrasonic time as influencing factors,Flavonoids yield as the evaluation index,the orthogonal experiment design method to optimize the extraction process.By colorimetric determination of total flavonoids of hydroxyl free radicals,superoxide anion scavenging ability.Results:The optimum extraction parameters were the solid-liquid ratio of 1∶20,ultrasonic power of 200 W,ultrasonic time of 50 min,and total flavonoids yield of 1.16%.There was an obvious dose-dependent relationship between the scavenging rate and the concentration of total flavone.The scavenging rates of 2.5μg/m L total flavone were 43.8%and 41.4%,respectively,with the strongest scavenging effect on hydroxyl radical and superoxide anion.Conclusion:This method is stable,feasible and reproducible,and can be used to extract total flavonoids of grapefruit peel with obvious antioxidant activity in vitro by ionic liquid,which can provide theoretical guidance for the amplification experiment or large-scale production of this system.
