基于网络药理学探讨益智仁对阿尔茨海默病的作用机制 被引量:4

Exploration on the Action Mechanism of Sharpleaf Galangal Fruit on Alzheimer’s Disease Based on Network Pharmacology

摘要 目的利用网络药理学的方法研究益智仁治疗阿尔茨海默病(AD)的作用机制。方法采用文献检索和数据库分析的方法,构建AD治疗相关靶点蛋白互作网络,运用DAVID数据库对相关靶点进行Gene Ontology(GO)功能分析及KEGG通路富集分析,并用Cytoscape ClueGo插件进行生物学功能分析。结果益智仁活性成分共13个,潜在作用靶点97个,运用CytoHubba筛出蛋白相互作用(PPI)关键靶点28个,GO生物功能分析共主要包括胶质细胞分化、神经炎症反应等,KEGG富集主要包括AGE-RAGE信号通路、IL-17信号通路等。结论益智仁可能是通过白杨素、豆甾醇、原儿茶酸等活性成分共同作用APP、JUN等靶点,生物学过程中星形胶质细胞活化促进靶点在AGE-RAGE通路或IL-17通路的表达来发挥改善AD,为益智仁对治疗AD的进一步研究提供参考。 Objective This paper aims to study action mechanism of Sharpleaf Galangal fruit upon Alzheimer’s disease(AD)with network pharmacology.Methods This paper constructed a complete AD-treatment-related targeted-protein interacting network with Documentation Retrieval and Database Analysis.And with combination of DAVID Database,it conducted a series of GO function analyses and KEGG enrichment analyses upon related target spots.And based on that,with Cytoscape ClueGo Plug-in,necessary biological-function analyses were made therein.Results It’s found that there are totally 13 active ingredients in Sharpleaf Galangal Fruit,including 97 potential targets.And with adoption of CytoHubba,totally 28 key PPI-related targets were successfully screened out.And the above GO biological-function analyses mainly focused on Glial-cell differentiation,neuroinflammation response and others.And KEGG enrichment ones mainly covered AGE-RAGE signaling pathway,IL-17 signaling pathway and etc.Conclusion Sharpleaf Galangal fruit may act on APP,JUN and other target spots via various active ingredients therein,such as Chrysin,Stigmasterol and PCA.In practical biological process,astrocytes were activated to promote expressions of these target spots in AGE-RAGE Pathway or IL-17 Pathway for purpose of further improving AD,as well as providing certain references for further researches of Sharpleaf Galangal fruit applied in treatments of AD.
