miR-106b-3p和miR-214在食管鳞癌组织中的表达及其与临床病理特征的关系

摘要 目的探讨miR-106b-3p和miR-214在食管鳞癌(ESCC)组织中的表达,分析其与患者临床病理特征的关系。方法选取2016年10月~2018年8月邯郸市第一医院确诊的行外科手术切除的ESCC患者60例,取其切除癌组织标本及癌旁组织标本,采用RT-qPCR技术检测ESCC患者癌组织和癌旁组织中miR-106b-3p及miR-214的表达水平,比较两者在癌组织和癌旁组织的表达差异,分析其与患者临床病理特征的关系及诊断ESCC的诊断。结果ESCC癌组织中miR-106b-3p和miR-214的表达量均高于癌旁组织,差异有统计学意义(P<0.05);不同淋巴结转移情况、肿瘤TNM分期的ESCC患者癌组织中miR-106b-3p、miR-214的表达比较,差异有统计学意义(P<0.05),不同年龄、性别的ESCC患者癌组织中miR-106b-3p、miR-214的表达比较,差异无统计学意义(P>0.05);ROC曲线显示,miR-106b-3p和miR-214诊断ESCC的敏感性分别为87.50%、85.00%,特异性分别为81.30%、87.50%。结论miR-106b-3p和miR-214在食管鳞癌组织中表达上调,可作为致癌基因参与食管鳞癌的发生发展,且两者对诊断该病具有一定的价值。 Objective To explore the expression of miR-106b-3p and miR-214 in esophageal squamous cell carcinoma(ESCC)tissues,and analyze their relationship with the clinical pathological characteristics of patients.Methods A total of 60 ESCC patients diagnosed with surgical resection in Handan First Hospital from October 2016 to August 2018 were selected,and their cancerous tissue specimens and para-cancerous tissue specimens were collected.RT-qPCR technology was used to detect the expression levels of miR-106b-3p and miR-214 in cancer tissues and adjacent tissues of ESCC patients.Compare the expression differences between the two in cancer tissues and adjacent tissues,analyze their relationship with the clinical pathological characteristics of patients,and diagnose ESCC.Results The expressions of miR-106b-3p and miR-214 in ESCC cancer tissues were higher than those in adjacent tissues,the difference was statistically significant(P<0.05);Comparison of the expression of miR-106b-3p and miR-214 in cancer tissues of ESCC patients with different lymph node metastasis and tumor TNM staging,the difference was statistically significant(P<0.05),There was no statistically significant difference in the expression of miR-106b-3p and miR-214 in ESCC patients of different ages and genders(P>0.05);The ROC curve showed that the sensitivity of miR-106b-3p and miR-214 in diagnosing ESCC were 87.50%and 85.00%,and the specificity was 81.30%and 87.50%,respectively.Conclusion miR-106b-3p and miR-214 are up-regulated in esophageal squamous cell carcinoma tissues,and can be used as oncogenes to participate in the occurrence and development of esophageal squamous cell carcinoma,and both have certain value in the diagnosis of the disease.
