猪伪狂犬病病毒安徽分离株对小鼠的致病性研究 被引量:2

摘要 为了解猪伪狂犬病病毒(PRV)安徽分离株的毒力特征,研究以小鼠为动物模型,对13株PRV安徽分离株(代号为AH1601~AH1604、AH1701~AH1704、AH1801~AH1805)进行半数致死量(LD50)测定、病理剖检及病理组织学观察、各组织脏器的病毒载量检测。结果显示,13株PRV对小鼠的LD50在10^2.569~10^5.167 TCID50之间,其中AH1802和AH1803的LD50最低,分别为10^2.569 TCID50和10^2.833 TCID50。病理剖检可见攻毒小鼠的肝、脾、肺、肾、脑等部位均有不同程度的出血、肿大,对照组无异常;病理组织HE检查13株PRV引起小鼠各组织脏器的病变存在差异,以AH1802和AH1803引起的病变最为严重。13株PRV安徽分离株在小鼠各组织脏器中的病毒载量存在差异,其中AH1802和AH1803在脑、肝脏、脾脏中的病毒载量显著高于其他11株(P<0.05),AH1802和AH1703在肺脏中的病毒载量显著高于其他11株(P<0.05),AH1601在肾脏中的病毒载量最高,AH1703、AH1704和AH1804在脑和肺脏中的病毒载量显著高于肝脏、脾脏和肾脏。结果表明,13株PRV安徽分离株对小鼠均具有较强的致病性,但来源于不同地区的各毒株之间对小鼠的致病力呈现不同。此研究丰富了安徽猪伪狂犬病流行病学资料,为深入研究PRV安徽分离株的抗原性奠定基础。 In order to understand the virulence characteristics of 13 PRV Anhui strains(code:AH1601~AH1604,AH1701~AH1704,AH1801~AH1805),mice were used as experimental animal models for LD50 determination,pathological dissection,histopathological observation and viral load detection of various tissues and organs.The results showed that the LD50 of 13 PRV strains to mice was 10^2.569~10^5.167 TCID50,the LD50 of AH1802 and AH1803 were the lowest,10^2.569 TCID50 and 10^2.833 TCID50 respectively.Clinical necropsy showed that the liver,spleen,lung,kidney,brain and other parts of the challenge group had different degrees of hemorrhage and swelling,and there was no abnormality in the control group;pathological tissue HE examination showed that 13 PRV strains caused different pathological changes in various tissues and organs of the mice,and the pathological changes caused by AH1802 and AH1803 were the most serious.The viral load of 13 PRV strains in various organs of mice was different.The viral load of AH1802 and AH1803 in brain,liver and spleen was significantly higher than that of other 11 strains(P<0.05).The viral load of AH1802 and AH1703 in lung was significantly higher than that of other 11 strains(P<0.05),AH1601 in kidney was the highest,AH1703,AH1704 and AH1804 in brain and lung were significantly higher than that of liver,spleen and kidney.The results showed that 13 PRV strains from Anhui Province had strong pathogenicity to mice,but the virulence of PRV strains varies from region to region.This study enriched the epidemiological data of PR in Anhui Province,and laid a foundation for further study on the antigenicity of PRV Anhui strains.
