结肠腺癌中eIF2c的表达及与细胞凋亡的关系 被引量:4

Expression of eukaryotic translation initiation factor 2c and its relationship with apoptosis in colorectal adenocarcinoma

摘要 目的 :检测结肠腺癌组织中真核翻译起始因子2c(eukaryotic translation initiation factor 2c,eIF2c)的表达,分析其与细胞凋亡的关系。方法:以69例结肠腺癌患者为研究对象,留取肿瘤组织为观察组,留取正常结肠黏膜组织为对照组。应用免疫组化法检测e IF2c、半胱氨酸肽酶-3(Caspase-3)的表达,应用Western blot检测B细胞淋巴瘤-2相关X蛋白(Bcl-2 associated X protein,BAX)的表达。构建过表达eIF2c结肠癌SW480细胞系,应用Western blot检测BAX的表达。结果:2组中eIF2c的阳性率差别有统计学意义(56.5%vs. 13.0%,χ^2=28.750,P=0.000)。eIF2c在不同肿瘤最大径(68.1%vs. 31.8%,χ^2=8.021,P=0.005)、浸润深度(81.2%vs. 35.1%%,χ^2=14.849,P=0.001)、有无坏死(48.0%vs. 78.9%%,χ^2=5.366,P=0.021)、淋巴结转移(83.3%vs.42.2%,χ^2=10.765,P=0.001)和癌栓(87.5%vs. 47.2%,χ^2=8.134,P=0.004)的分组中,差异有统计学意义。e IF2c与Caspase-3呈负相关性(r=-0.69,P=0.000),eIF2c与BAX呈负相关性(r=-0.66,P=0.000)。生存分析显示,eIF2c与生存时间相关(χ^2=14.00,P=0.000)。过表达e IF2c结肠癌细胞系中BAX表达下降。结论:e IF2c在结肠腺癌中高表达,与临床病理特征和预后相关,e IF2c异常表达与凋亡因子有关联。 Objective:To detect the expression of eukaryotic translation initiation factor 2 c(e IF2 c)in colorectal adenocarcinoma and to analyze its relationship with apoptosis.Methods:A total of 69 patients with colorectal adenocarcinoma were taken as the study object.Tumor tissue was taken as the observation group and normal colonic mucosa tissue was taken as the control group.The expressions of eIF2 c and Caspase-3 were detected by immunohistochemistry method.Expression of Bcl-2 associated X protein(BAX)was detected by Western blot.Colon cancer SW480 cell line of overexpression eIF2 c was constructed and expression of BAX was detected by Western blot.Results:Differences of positive rate in two groups was statistically significant(56.5%vs.13.0%,χ^2=28.750,P=0.000).Expression of eIF2 c was significantly different in maximum diameter(68.1%vs.31.8%,χ^2=8.021,P=0.005),invasion depth(81.2%vs.35.1%,χ^2=14.849,P=0.001),necrosis(48.0%vs.78.9%,χ^2=5.366,P=0.021),lymph node metastasis(83.3%vs.42.2%,χ^2=10.765,P=0.001)and tumor thrombus(87.5%vs.47.2%,χ^2=8.134,P=0.004).Negative correlation was found between eIF2 c and Caspase-3(r=-0.69,P=0.000),and eIF2 c and Bax(r=-0.66,P=0.000).According to survival analysis,e IF2 c was related with survival time(χ^2=14.00,P=0.000).Expression of BAX was decreased in colon cancer cell line of overexpressed eIF2 c.Conclusion:Higher expression of eIF2 c is related with clinicopathological features and prognosis in colorectal adenocarcinoma.Abnormal expression of eIF2 c is related with apoptosis factors.
