摘要 目的:建立同时测定七味广枣散中没食子酸、5-羟甲基糠醛、原儿茶酸、丁香酚含量的方法。方法:采用HPLC法,SHIMADZU Shim-pack GIST(250 mm×4.6 mm,5μm)色谱柱,以乙腈-0.5%乙酸水溶液为流动相,梯度洗脱,流速为1.0 mL/min,检测波长为254 nm,柱温为30℃,进样量为10μL。结果:没食子酸、5-羟甲基糠醛、原儿茶酸、丁香酚分别在2.42~48.4μg/mL(r=0.999 9)、2.88~57.6μg/mL(r=0.999 7)、1.12~22.4μg/mL(r=1)、102.0~2040μg/mL(r=0.999 9)浓度范围内与峰面积呈良好的线性关系;平均加样回收率分别为99.57%、98.22%、101.35%、96.59%,RSD分别为0.99%、1.04%、1.53%、0.20%(n=6)。结论:该方法简便、快速、准确、分离效果好,可为七味广枣散的质量评价和控制提供参考。 Objective: To establish a method for the simultaneous determination of gallic acid, 5-hydroxymethylfurfural, protocatechuic acid and eugenol in Qiwei Guangzao Powder(七味广枣散). Methods: HPLC was performed on SHIMADZU Shim-pack GIST(250 mm×4.6 mm, 5 μm) column with acetonitrile-0.5% acetic acid as mobile phase, gradient elution, flow rate of 1.0 mL/min, detection wavelength of 254 nm, column temperature of 30℃ and injection volume of 10 μL. Results: The concentrations of gallic acid, 5-hydroxymethylfurfural, protocatechuic acid and eugenol were 2.42-48.4 μg/mL(r=0.999 9), 2.88-57.6 μg/mL(r=0.999 7), 1.12-22.4 μg/mL(r=1)and 102.0-2 040 μg/mL(r =0.999 9), respectively. The average recoveries were 99.57%, 98.22%, 101.35%96.59%, with RSD of 0.99%, 1.04%, 1.53% and 0.20%(n=6), respectively. Conclusion: The method is simple,rapid, accurate, and has good separation effect. It can provide a reference for the quality evaluation and control of Qiwei Guangzao Powder.
