摘要
利用Box-Benhnken响应面法对热水浸提羊肚菌多糖工艺进行优化,在单因素实验基础上,选择水料比、提取温度、提取时间为自变量,以羊肚菌多糖提取率为响应值,采用三因素三水平的响应面分析法优化羊肚菌多糖提取工艺.结果表明,影响热水浸提羊肚菌多糖提取率的大小顺序为:提取温度>提取时间>水料比.羊肚菌多糖提取的最佳工艺条件为:水料比51∶1(mL/g)、提取温度46℃、提取时间3.5 h,平均提取率为7.39%(n=3),与预测值7.311%接近.
Box-Benhnken response surface methodology was used to optimize the hot water extraction process of morchella esculenta polysaccharide.On the basis of single factor experiment,the water-material ratio,extraction temperature and extraction time were selected as independent variables,the extraction rate of morchella esculenta polysaccharide was taken as response value,and the extraction process of morchella esculenta polysaccharide was optimized by response surface analysis with 3 factors and 3 levels.The results showed that the order of factors affecting the hot water extraction rate of Morchella polysaccharide was as follows:extraction temperature>extraction time>water-material.The optimum extraction conditions of morchella esculenta polysaccharide were:water/material ratio 51∶1(mL/g),extraction temperature 46℃,and extraction time 3.5 h,with an average extraction rate of 7.39%(n=3),which was close to the predicted value of 7.311%.
