高效液相色谱法测定不同产地麸炒白术中苍术酮、白术内酯Ⅰ、白术内酯Ⅱ和白术内酯Ⅲ的含量 被引量:13

摘要 目的采用高效液相色谱法测定不同产地麸炒白术中苍术酮、白术内酯Ⅰ、白术内酯Ⅱ和白术内酯Ⅲ的含量,对不同产地麸炒白术进行质量评价。方法色谱柱为Dikma Diamonsil C8(150 mm×4.6 mm,5μm),流动相为水-乙腈(梯度洗脱),流速1.0 mL/min,检测波长为220和275 nm,柱温30℃,进样量20μL。结果在上述色谱条件下,苍术酮、白术内酯Ⅰ、白术内酯Ⅱ和白术内酯Ⅲ浓度分别在47.600~142.800、1.304~3.912、1.126~3.378、1.740~5.220 mg/L内线性关系良好,平均加样回收率分别为99.98%、99.63%、99.15%、99.79%,相对标准偏差(RSD)分别为0.03%、1.70%、1.57%、1.10%。结论该方法分离度好、专属性强、重现性好、简单易行,可作为不同产地麸炒白术饮片的质量控制方法;测定结果表明,浙江磐安产麸炒白术饮片中4种成分含量相对较高。 Objective To determine the content of Atractylone,AtractylenolideⅠ,AtractylenolideⅡand AtractylenolideⅢin Atractylodis Macrocephalae Rhizoma stir-fried with wheat bran from different habitats by high performance liquid chromatogram method,and to evaluate the quality of Atractylodis Macrocephalae Rhizoma stir-fried with wheat bran from different habitats.Methods The separation was performed on Dikma Diamonsil C8(150 mm×4.6 mm,5μm)column with mobile phase consisted of water-acetonitrile(gradient elution)at the flow rate of 1.0 mL/min.The detection wavelength was set at 220 and 275 nm,the column temperature was 30℃and injection volume was 20μL.Results Under the above chromatographic conditions,Atractylone,AtractylenolideⅠ,AtractylenolideⅡand AtractylenolideⅢshowed good linear relationship in the ranges of 47.600-142.800,1.304-3.912,1.126-3.378,1.740-5.220 mg/L.The average recovery was 99.98%,99.63%,99.15%,99.79%,respectively,and the relative standard deviation(RSD)was 0.03%,1.70%,1.57%,1.10%,respectively.Conclusion This method is well-separated,specific,reproducible,simple and feasible.It can be used for the quality control method of Atractylodis Macrocephalae Rhizoma stir-fried with wheat bran.The result of determination shows that the content of the four components in Atractylodis Macrocephalae Rhizoma stir-fried with wheat bran from Zhejiang Pan′an is relatively higher.
