摘要
试验通过对阿克苏某生猪养殖场腹泻仔猪肛拭子进行病原分离,并成功分离到一株细菌。细菌纯化后通过革兰氏染色观察其形态结构,并提取其DNA测序。分离菌株得到的基因序列标记为W1,通过DNA star生物软件将菌株W1与国内外菌株进行基因序列对比,并做同源性和进化树分析。结果表明:W1与国内外15株奇异变形杆菌的同源性为96.4%~99.4%,其中与W1同源性最低的是KT216564,与W1同源性最高的是AB272366。遗传进化树分析表明,分离株W1与南京株JF775415处于同一分支,属于奇异变形杆菌,该菌株的成功分离为阿克苏地区对奇异变形杆菌的研究提供参考。
In the experiment, the pathogen was isolated from anal swabs of diarrhea piglets from a pig farm in Aksu, and a strain of bacteria was successfully isolated. The isolated bacteria were purified and observed their basic morphological structure by gram staining, and their DNA was extracted for sequencing. The gene sequence obtained by the isolated strain is marked as W1, and the gene sequence of the strain W1 was compared with the domestic and foreign strains through the DNA star biological software, and the homology and evolutionary tree analysis are performed. The test results showed that the homology between W1 and 15 strains of Proteus mirabilis at home and abroad is 96.4%~99.4%, of which KT216564 had the lowest homology with W1 and AB272366 had the highest homology with W1. The analysis of the genetic tree, the isolate W1 was in the same branch as the Nanjing strain JF775415 and belongs to Proteus mirabilis. The successful isolation of this strain provides a reference for the study of Proteus mirabilis in Aksu area.
