摘要
目的:探索乳腺癌细胞系MCF-7与巨噬细胞半乳糖型凝集素(MGL)的结合情况,并对MCF-7细胞表面的MGL配体进行鉴定。方法:采用流式细胞术和单糖抑制实验检测MCF-7细胞表面糖抗原的GalNAc表达以及MGL与MCF-7细胞的结合情况。RT-PCR、Western blot和免疫沉淀等研究方法对MCF-7上MGL配体进行鉴定。结果:MGL能够与乳腺癌细胞MCF-7高度结合,且这种结合是Ca2+依赖的;乳腺癌细胞MCF-7表面高表达GalNAc,即Tn抗原;单糖GalNAc的加入可明显抑制MGL与乳腺癌细胞MCF-7的相互作用;黏蛋白MUC1为乳腺癌细胞系MCF-7表面MGL的配体。结论:黏蛋白MUC1为乳腺癌细胞系MCF-7表面MGL的配体,介导了MGL对乳腺癌细胞的免疫识别。
Objective:To investigate the binding of breast cancer cell line MCF-7 with macrophage galactose lectin(MGL),and to identify the MGL ligand on the surface of MCF-7 cell.Methods:Flow cytometry and monosaccharide inhibition test were used to detect the expression of GalNAc on the surface of MCF-7 cells,and the binding of MGL to MCF-7 cells.The MGL ligands on MCF-7 were identified by RT-PCR,Western blot and immunoprecipitation.Results:MGL could highly bind to MCF-7 cells,and the binding was Ca2+-dependent;the surface of MCF-7 cells highly expressed GalNAc,namely Tn antigen.The addition of monosaccharide GalNAc could significantly inhibit the interaction between MGL and MCF-7.Mucin MUC1 was the ligand of MGL on MCF-7.Conclusion:MUC1 is a ligand of MGL on the surface of MCF-7 cell line and mediates the immune recognition of breast cancer cells by MGL.
