基于TCGA数据库研究NUF2基因在肝细胞癌中的表达及临床意义

Study of expression and clinical significance of NUF2 gene in hepatocellular carcinoma based on TCGA database

目的:通过下载TCGA数据库中肝细胞癌(HCC)数据探究NUF2基因在HCC中的表达及临床意义。方法:从TCGA数据库下载正常肝组织和肿瘤组织测序数据及临床信息资料,采用R语言等软件分析NUF2基因的表达差异及其与临床病理特征的关系,单因素和多因素Cox回归模型分析HCC发生发展相关风险因素,GSEA软件预测NUF2基因可能的信号通路。结果:HCC组织中NUF2表达显著高于正常组织(P<0.05),且和年龄、临床分期、肿瘤分级及T分期显著相关,单、多因素Cox分析表明NUF2基因可作为HCC的独立预后因素(P<0.05)。GSEA结果显示NUF2高表达与肿瘤相关通路显著相关。结论:HCC组织中NUF2基因高表达且和病理特征相关,可能是HCC诊断和治疗的潜在生物学标记物与治疗靶点。

Objective:To investigate expression and clinical significance of NUF2 in hepatocellular carcinoma(HCC)on basis of data from TCGA database.Methods:RNA-seq and clinical information in HCC tissues and nornal tissues were downloaded from TCGA database,expression difference of NUF2 and its relationship with clinicopathological characteristics were analyzed by R software.Univariate and multivariate Cox regression model to analyze relevant risk factors of HCC occurrence and development.Signal pathways of NUF2 gene were analyzed by GSEA software.Results:Expression level of NUF2 in HCC tissues was significantly higher than that in normal tissues(P<0.05),which was significantly correlated with age,stage,grade and T classification.Univariate and multivariate Cox regression analysis showed that NUF2 was independent prognostic factor of HCC(P<0.05).GSEA results revealed that high expression of NUF2 significantly correlate with tumor-related pathways.Conclusion:NUF2 gene expression is significantly upregulated in HCC tissue and highly associated with clinicopathological characteristics,which could be a potential candidate biomarker and therapeutic target for HCC.
