调治天癸方对雷公藤多苷致少弱精症大鼠精子质量及Fas表达的影响研究 被引量:2

摘要 目的探讨调治天癸方对少弱精症大鼠精子质量及睾丸组织中Fas表达的影响。方法将72只SD雄性大鼠随机分为正常组、模型组、生精胶囊组和调治天癸方高、中、低剂量组,每组12只。除正常组给予正常饮食外,其余各组大鼠连续灌胃雷公藤多苷片20 mg/(kg·d)5周建立少弱精症模型。造模成功后,生精胶囊组给予生精胶囊粉末0.06 g/100 g灌胃,调治天癸方高、中、低剂量组分别给予调治天癸方水煎液3.38 g/100 g、1.69 g/100 g、0.85 g/100 g灌胃,正常组及模型组给予等量生理盐水灌胃,均连续灌胃5周。末次灌胃后取大鼠双侧睾丸及附睾,采用计算机辅助精子分析方法检测精子密度及活动率,Western blot法检测睾丸组织中Fas蛋白表达情况。结果与正常组比较,模型组大鼠精子密度、精子活动率均明显降低(P均<0.05),睾丸组织中Fas蛋白表达水平明显升高(P<0.05);而生精胶囊组及调治天癸方各剂量组大鼠精子密度、活动率均明显高于模型组(P均<0.05),睾丸组织中Fas蛋白表达水平均明显低于模型组(P均<0.05);且调治天癸方各剂量组大鼠精子密度、活动率均明显高于生精胶囊组(P均<0.05),睾丸组织中Fas蛋白表达水平均明显低于生精胶囊组(P均<0.05),且调治天癸方高、中、低剂量组Fas蛋白表达水平呈剂量依赖性降低。结论调治天癸方可明显改善雷公藤多苷致少弱精症大鼠的精子质量,对睾丸组织中Fas的表达有明显抑制作用。 Objective It is to explore the effect of Decoction for regulating Tiangui(DRT)on sperm quality and Fas expression in rats with oligoasthenospermia induced by tripterygium glycoside.Methods Seventy-two SD male rats were randomly divided into normal group,model group,Shengjing capsule group and DRT high dose group,medium dose group and low dose group,12 rats in each group.The rats in the normal group were given normal diet,the rats in the other groups were treated with tripterygium glycoside 20 mg/(kg·d)by gavage for 5 weeks to establish oligoasthenospermia models.After successful modeling,Shengjing capsule group was given Shengjing capsule powder 0.06 g/100 g by gavage,the DRT high,medium and low dose groups were given DRT water decoction 3.38 g/100 g,1.69 g/100 g,0.85 g/100 g by gavage respectively,and the normal group and the model group were given the same amount of normal saline by gavage,all the groups were continuously treated for 5 week.After the last gavage,the rat’s bilateral testes and epididymis were taken,computer-aided sperm analysis was used to detect sperm density and motility,and Western blot was used to detect the expression of Fas protein in testicular tissue.Results Compared with the normal group,the sperm density and sperm motility rate of the model group were significantly reduced(all P<0.05),and the Fas protein expression level in the testicular tissue was significantly increased(P<0.05);the sperm density and motility rate of rats in the Shengjing capsule group and each DRT dose group were significantly higher than those in the model group(all P<0.05),and the Fas protein expression in testicular tissue was significantly lower than that in the model group(all P<0.05);the sperm density and motility rate of rats in each DRT dose group were significantly higher than those in the Shengjing capsule group(all P<0.05),and the Fas protein expression in testicular tissue was significantly lower than that in the Shengjing capsule group(all P<0.05),the expression level of Fas protein decreased in a dose-dependent manner in every DRT dose group.Conclusion DRT can significantly improve the sperm quality of rats with oligoasthenospermia caused by tripterygium polyglycosides,and it has a significant inhibitory effect on the expression of Fas in testicular tissues.
