补虚解毒化瘀方对单侧输尿管梗阻大鼠健侧肾脏肾小球细胞增殖的抑制作用 被引量:1

摘要 目的观察补虚解毒化瘀方对单侧输尿管梗阻大鼠健侧肾脏肾小球细胞增殖的抑制作用。方法结扎大鼠单侧输尿管诱导肾脏损伤。40只大鼠随机分为假手术组、模型组、依普利酮组和中药治疗组。10 d后苏木精-伊红染色(HE)观察肾脏组织病理变化;Sirius red染色检测胶原在肾脏的沉积;免疫组织化学染色及蛋白印迹方法检测缺氧诱导因子1α(HIF-1α)、转化生长因子(TGF-β1)和增殖细胞核抗原(PCNA)的表达。结果模型组较假手术组肾小球PCNA阳性细胞明显增多(P<0.05),肾组织中HIF-1α、TGF-β1、PCNA蛋白表达明显升高(P<0.05);补虚解毒化瘀方及醛固酮受体阻断剂依普利酮可以下调其表达(P<0.05)。结论补虚解毒化瘀方可通过调节HIF-1α的表达抑制单侧输尿管梗阻健侧肾小球细胞增殖。 AIM To observe the effect of Buxu Jiedu Huayu Decoction on inhibiting glomerular cell proliferation in contralateral kidneys of unilateral ureteral obstruction rats.METHODS The rats renal injuries were induced by unilateral ureteral obstruction.Forty rats were randomly assigned into sham operation group,unilateral ureteral obstruction group,eplerenone group and Buxu Jiedu Huayu Decoction group.Ten days after the corresponding intervention,the rats had their renal pathological changes and renal fibrosis detected respectively by hematoxylin-eosin staining(HE)and Sirius red staining.In addition,the expression of HIF-1α,TGF-β1 and PCNA were measured by Western blot and immunohistochemical staining.RESULTS Compared with the sham group,unilateral ureteral obstruction model group shared significantly increased PCNA positive cells,and significantly up-regulated expression of HIF-1α,TGF-β1 and PCNA in renal tissues(P<0.05)which can be offset by either Buxu Jiedu Huayu Decoction or eplerenone.CONCLUSION Buxu Jiedu Huayu Decoction inhibits the cell proliferation in contralateral kidneys of unilateral ureteral obstruction rats via down-regulating HIF-1αexpression.
