通阳化浊方干预冠心病的动物实验研究

Animal Experimental Study on Treatment of CHD by Tongyang Huazhuo Decoction

目的通过动物实验,探索通阳化浊方治疗冠心病的疗效与机制。方法 50只雄性白兔,随机分为空白组、模型组、中药阳性对照组、实验1组(通阳化浊方组)与实验2组(四妙勇安汤组) 5组,每组10只。空白组普通饲料喂养;模型组高脂饲料喂养至8周,并于第4周后采用球囊手术损伤雄性白兔右侧颈总动脉;中药阳性对照组和两个实验组在模型组基础上,分别在术后给予血脂康胶囊、通阳化浊方和四妙勇安汤灌胃,每日1次,连续4周。给药结束后,收集血液、颈总动脉血管组织和新鲜粪便进行检测。结果与模型组比较,通阳化浊方组与四妙勇安汤组均能降低血脂、循环内皮细胞和ET浓度,提升NO浓度,改善血管病理形态,并且调整肠道菌群。结论通阳化浊方对于冠心病模型实验兔治疗有效;通阳化浊汤与四妙勇安汤可从不同角度和途径改善冠心病。

Objective Through Tongyang Huazhuo Decoction experiment,we explored its efficacy and mechanism in coronary atherosclerotic heart disease.Methods The experimental animals were studied by 50 male white rabbits.They were randomly divided into 5 groups: blank group,model group,Chinese medicine positive group,Tongyang Huazhuo Decoction group and Simiao Yong’an Decoction group,with 10 rabbits in each group.The blank group and model group were fed with normal diet and high fat diet for 8 weeks.The model group made right carotid artery injury surgery with balloon in the 4 thweek.On the basis of the model group,Chinese medicine positive group and two test groups were fed with Xuezhikang Capsule,Tongyang Huazhuo Decoction and Simiao Yong’an Decoction orally once a day,for 4 consecutive weeks.After treatment,male rabbits were anesthetized to collect blood,carotid artery endothelial tissue and fresh dejection.Results Compared with the model group,both Tongyang Huazhuo Decoction and Simiao Yong’ an Decoction groups could reduce blood lipid,circulating endothelial cells and ET concentration,increase NO concentration,improve vascular pathological morphology,and regulate intestinal flora.Conclusion Tongyang Huazhuo Decoction is effective in the treatment of coronary atherosclerotic heart disease model rabbits.Tongyang Huazhuo Decoction and Simiao Yong’an Decoction can improve coronary heart disease from different angles and approaches.
