赤胫散指纹图谱的建立及其3种化学成分含量的测定 被引量:1

摘要 目的建立赤胫散指纹图谱的定性分析和没食子酸、鞣花酸及3,3’-二甲基鞣花酸成分定量分析的测定方法。方法选取本省野生赤胫散药材17批次,采用高效液相色谱法(HPLC)建立赤胫散指纹图谱,采用相似度分析、聚类分析、主成分分析及正交偏最小二乘判别分析进行质量评价,并测定3种化学成分含量。结果建立了赤胫散的HPLC指纹图谱共有模式,确定了19个共有峰,17批次赤胫散的指纹图谱相似度为0.768~0.997,可分为2类;没食子酸、鞣花酸及3,3’-二甲基鞣花酸的含量,分别在1.23~39.20 mg/L(r=0.999 8)、7.98~255.30 mg/L(r=0.999 8)及0.90~28.80 mg/L(r=0.999 9)的进样浓度范围内线性关系良好,加样回收率为96.90%~103.48%,相对标准偏差(RSD)<3%。结论建立的赤胫散指纹图谱和含量测定方法准确可靠,可用于赤胫散的质量控制。 Objective To establish the qualitative analysis of the fingerprint spectrum of Polygonum runcinatum( P. runcinatum) and the quantitative analysis of gallic acid,ellagic acid and 3,3 ’-dimethylellagic acid. Methods Seventeen batches of wild P. runcinatum from Guizhou province were selected. High performance liquid chromatography( HPLC) was used to establish the fingerprint of P.runcinatum. Similarity analysis,cluster analysis,principal component analysis( PCA) and orthogonal partial least squares discriminant analysis were used to assess the quality. Three chemical components of P. runcinatum were determined. Results The HPLC fingerprint of the P. runcinatum was established.Nineteen identified peaks were common. The fingerprint similarities in 17 batches of P. runcinatum was ranged from 0. 768 ~ 0. 997. Cluster analysis showed that 17 batches belonged two clusters.Within the range of injection concentration,the concentrations of gallic acid,ellagic acid and 3,3 ’-dimethyl ellagic acid were 1. 23 ~ 39. 20 mg/L( r = 0. 999 8),7. 98 ~ 255. 30 mg/L( r = 0. 999 8)and 0. 90 ~ 28. 80 mg/L( r = 0. 999 9),respectively,showing a good linear relationship. The recovery rate of injection is 96. 90% ~ 103. 48% with the relative standard deviation( RSD) < 3%.Conclusion The established fingerprint spectrum and composition determination of P. runcinatum are accurate and reliable,and can be used for quality control of P. runcinatum.
