摘要
细胞内黏度和过氧亚硝酸根离子(ONOO^(-))含量影响正常细胞代谢和生理功能,与很多疾病过程密切相关。为此,设计合成了一种新型黏度和ONOO^(-)双响应荧光探针VI-PN(2-((6-羟基-2,3-二氢-1H-氧杂蒽-4-基)亚甲基)丙二腈)。在溶液体系中,该探针随着溶液(PBS/甘油)体系黏度增大,红色荧光(λem1=634 nm)显著增强,荧光增强倍数(F/F0)为14倍;当加入ONOO^(-)后,探针被氧化,形成一种新的荧光团,产生绿色荧光(λem2=559 nm),荧光增强倍数为240倍。该探针细胞毒性低,对黏度和ONOO^(-)的响应具有选择性好和响应灵敏的特点,可区分和定性地检测黏度和ONOO^(-)的变化,并应用于细胞内黏度和ONOO^(-)浓度的共聚焦荧光成像检测。
Intracellular viscosity and peroxynitrite(ONOO^(-))influences normal cell metabolism and function processes,associated with a large number of diseases.Herein,a novel viscosity and ONOO^(-)dual-responsive fluorescent probe VI-PN(2-((6-hydroxy-2,3-dihydro-1H-xanthen-4-yl)methylene)malononitrile)was designed and prepared.In a mixture of PBS/glycerol solution,the probe displayed remarkable red fluorescence emission(λem1=634 nm)increase with the increasing viscosity and a largest fluorescence enhancement factor(F/F0)of 14-fold.Upon the addition of ONOO^(-),the probe was oxidized to produce another fluorophore and displayed significant green fluorescence emission(λem2=559 nm)with a largest fluorescence enhancement factor of 240-fold.Thereby,the probe,featured with low cytotoxicity,can well distinguish and determine viscosity and ONOO^(-) in buffer solution,respectively.As a result,the probe can monitor the content change of viscosity and ONOO^(-) in live cells by confocal fluorescence imaging.
