摘要
目的探讨碱性成纤维细胞生长因子(Basic Fibroblast Growth Factor,bFGF)联合胶原膜对大鼠硬腭软组织缺损病理及创面愈合的影响。方法采用30只SPF级健康Wistar大鼠建立硬腭软组织缺损模型,随机分为3组:Bf组、Dd组、Nc组,每组各10只。Bf组大鼠采用结合bFGF/胶原膜修复创面,Dd组仅采用胶原膜修复,Nc组不进行修复处理,在术后2周处死大鼠,切取硬腭组织制成切片,HE染色观察组织病理,计算创面愈合率,观察新生微血管数量,采用RT-PCR及Westernblot法检测Ⅰ型胶原(TypeⅠcollagen,ColⅠ)、ColⅢ水平表达。结果HE染色观察可见,Bf组可发现成纤维细胞生成增多,明显新生血管生成,仅见少量胶原膜,Dd组新生血管数量及成纤维细胞较Bf组少,Nc组新生血管数量及成纤维细胞少于其他两组;Bf组创面愈合率为(87.65±5.03)%,显著高于Dd组(81.03±3.98)%与Nc组(72.12±3.16)%,3组比较差异有统计学意义(P<0.05);Bf组新生血管数量为(8.96±0.78),高于Dd组(6.23±0.71)与Nc组(4.29±0.69),3组比较差异有统计学意义(P<0.05);与Nc组比较,Dd组大鼠ColⅠ、ColⅢmRNA、蛋白表达量有所降低,与Dd组比较,Bf组大鼠ColⅠ、ColⅢmRNA、蛋白表达也有所降低,上述差异均有统计学意义(P<0.05)。结论bFGF联合胶原膜能进一步促进硬腭软组织缺损的修复,提高创面愈合率,该作用可能与下调ColⅠ、ColⅢ表达有关。
Objective To explore the effect of bFGF combined with collagen film on pathology and wound healing of hard palate soft tissue defects in rats.Methods Thirty SPF healthy Wistar rats were used to establish a soft palate soft tissue defect model,which was divided into Bf group(n=10),Dd group(n=10)and Nc Group(n=10).Bf groupreceviedbFGF/collagen membrane,Dd group receviedcollagen membrane,Nc group was not repaired.The rats were killed 2 weeks after operation,and the hard palate tissue was cut into sections.The histopathology was used to observe by HE staining.Thewound healing rate and number of new microvessels were observed and the protein expressionof ColⅠand ColⅢwere detected by RT-PCR and Western-blot method.Results HE staining showed increasedtibroblastsand neovascular in Bf group,only a small amount of collagen membrane was seen.The number of neovascularization and fibroblasts in Dd group were less than those in Bfgroup,and the number of neovascular and fibroblasts in Ncgroup were less than those in the other two groups.The wound healing rate of the Bf group was the highest(87.65±5.03)%,followed by the Dd group(81.03±3.98)%,the wound healing rate in the Nc group was the lowest(72.12±3.16)%,and the differencein three groups was significant(P<0.05).The number of neovascular in the Bf group was the largest(8.96±0.78),followed by that in the Dd group(6.23±0.71),and the lowest in the Nc group(4.29±0.69),the difference in three groups was significant(P<0.05).Compared with the Nc group,the protein expression of ColⅠand ColⅢmRNA in the Dd group decreased significantly.Compared with the Dd group,the protein expression of ColⅠand ColⅢin the Bf group decreased,all difference was significant(P<0.05).Conclusion bFGF combined with collagen membrane can further promote the repair of hard palate soft tissue defects and improve the wound healing rate,which may be related to the down regulation of ColⅠand ColⅢexpression.
