基于生物信息数据库分析FEN1基因在乳腺癌中的表达及其临床意义 被引量:2

摘要 目的通过分析生物信息数据库相关资料,探讨瓣状核酸内切酶1(FEN1)基因在乳腺癌中的表达及其临床意义。方法检索Oncomine数据库,分析FEN1基因在不同类型肿瘤中的表达情况;检索基因表达谱动态分析(GEPIA)数据库,对比乳腺癌组织与正常乳腺组织之间FEN1基因的表达差异,分析该基因的表达水平与乳腺癌临床病理分期的关系;检索人类蛋白质图谱(HPA)数据库,可视化乳腺癌组织与正常乳腺组织FEN1基因的差异表达情况;利用Kaplan-Meier Plotter在线分析FEN1基因表达水平与乳腺癌患者预后的关系;检索肿瘤单细胞(CancerSEA)数据库分析FEN1基因与乳腺癌单个细胞功能状态的相关性。结果FEN1在多种肿瘤组织中高表达。与正常乳腺组织相比,乳腺癌组织中FEN1基因的表达明显升高(P<0.05),且不同临床病理分期乳腺癌患者FEN1基因表达水平比较差异有统计学意义(P<0.05)。从总体来看,FEN1基因高表达组总生存率低于低表达组(P<0.05)。此外,FEN1基因的表达与乳腺癌多种细胞功能状态相关,其中与乳腺癌细胞的细胞周期、DNA损伤、DNA修复正相关性最高。结论FEN1基因在乳腺癌组织中呈高表达,与患者预后相关,并且与乳腺癌细胞功能存在一定相关性,可为临床乳腺癌的治疗及基因靶向药物的研制提供重要的理论依据。 Objective To analyze the expression of flap endonuclease 1(FEN1)gene in breast cancer and its clinical significance based on bioinformatics database analysis.Methods The Oncomine database was searched to analyze the expression of FEN1 gene in different types of tumors.The gene expression profiling interactive analysis(GEPIA)database was used to compare the difference of FEN1 gene expression between breast cancer and normal tissues,and to analyze the relationship between the expression level of FEN1 gene and the clinicopathological stage of breast cancer.The differential expression of FEN1 gene between breast cancer tissues and normal tissues was visualized by searching human protein atlas(HPA)database.Kaplan-Meier Plotter was used to analyze the clinical significance of FEN1 gene expression.The correlation between FEN1 gene and functional states of single breast cancer cells was analyzed using CancerSEA database.Results FEN1 was highly expressed in many kinds of tumor tissues.The expression level of FEN1 gene in breast cancer tissues was significantly higher than that in normal tissues(P<0.05).There was significantly different in the expression of FEN1 gene among breast cancer patients with different clinicopathological stages.The survival time of the FEN1 gene in highly expressed group was significantly lower than that of the low expression group.In addition,the expression of FEN1 gene was correlated with a variety of functional states of breast cancer cells,among which the correlation of cell cycle,DNA damage and DNA repair was the highest in breast cancer cells.Conclusion FEN1 gene is highly expressed in breast cancer tissue,which is related to the prognosis of patients and has a certain correlation with the function of breast cancer cells,providing information for the clinical treatment of breast cancer and the development of gene-targeted drugs.
